Cefazolin is a first-generation cephalosporin antibiotic used to treat a variety of bacterial infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Typical oral dosage for moderate infections: 250 mg to 1 gram every 6-8 hours. For severe infections, dosage may be increased.
Elderly
Dosage adjustment may be required based on renal function, typically lower doses or extended dosing intervals.
Renal_impairment
Reduced dosage frequency or amount is necessary based on creatinine clearance. Consult a physician for specific guidelines.
How to Take

Mechanism of Action
Cefazolin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs) located in the bacterial cell wall, leading to cell lysis and death.

Excretion
Primarily excreted unchanged in the urine by glomerular filtration and tubular secretion. About 60-70% of the dose is excreted within 6 hours.
Half life
Approximately 1.8 hours in adults with normal renal function.
Absorption
Well absorbed from the gastrointestinal tract following oral administration. Peak plasma concentrations are achieved within 1-2 hours.

Side Effects
Contraindications
- •Known hypersensitivity to cefazolin, other cephalosporins, or any component of the formulation.
- •Previous severe hypersensitivity reaction to penicillin or other beta-lactam antibiotics.
Drug Interactions
Probenecid
Concomitant administration with probenecid decreases the renal tubular secretion of cefazolin, resulting in increased and prolonged plasma concentrations of cefazolin.
Oral anticoagulants (e.g., warfarin)
May enhance the anticoagulant effect by interfering with vitamin K synthesis in the gut. Monitor INR levels.
Nephrotoxic drugs (e.g., aminoglycosides, furosemide)
Concurrent use may increase the risk of nephrotoxicity. Monitor renal function closely.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric distress, and diarrhea. In severe cases, neurotoxicity (e.g., seizures) may occur. Treatment is symptomatic and supportive. Hemodialysis may be useful in accelerating the removal of cefazolin from the blood.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown a risk, but there are no adequate and well-controlled studies in pregnant women. Use only if clearly needed. Cefazolin is excreted in breast milk in low concentrations; use with caution in breastfeeding mothers.
