Description
Overview of the medicine
Apimox 125 mg Pediatric Drop is an antibiotic belonging to the penicillin group. It is used to treat a wide variety of bacterial infections in infants and young children, including infections of the ear, nose, throat, lower respiratory tract, urinary tract, and skin. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Not typically used in drop form for adults. Refer to tablet/capsule dosages. Typical Amoxicillin adult dose is 250-500 mg every 8 hours or 500-875 mg every 12 hours depending on infection severity.
Elderly
Similar to adult dosage with caution for renal impairment. Refer to tablet/capsule dosages.
Renal_impairment
Dosage adjustment is required for severe renal impairment (CrCl <30 mL/min). Reduce dose or extend dosing interval.
How to Take
Administer orally. Shake the reconstituted suspension well before each use. Can be given with or without food. Use the provided dropper for accurate dosing.
Mechanism of Action
Amoxicillin is a beta-lactam antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Oral: 30-60 minutes
Excretion
Primarily excreted unchanged by the kidneys via tubular secretion and glomerular filtration. About 60-80% of an oral dose is excreted in urine within 6 hours.
Half life
1-1.5 hours in adults with normal renal function; longer in neonates and those with renal impairment.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, especially in the presence of food. Bioavailability is approximately 70-90%.
Metabolism
Limited metabolism, primarily to inactive penicilloic acid.
Side Effects
Contraindications
- Hypersensitivity to amoxicillin, penicillin, or any other beta-lactam antibiotics.
- History of severe hypersensitivity reaction to another beta-lactam agent (e.g., cephalosporin, carbapenem, monobactam).
Drug Interactions
Probenecid
Increases and prolongs amoxicillin blood levels by decreasing renal tubular secretion.
Methotrexate
May increase methotrexate toxicity by inhibiting its renal excretion.
Oral contraceptives
May reduce efficacy of oral contraceptives (rare, but advise additional contraception).
Warfarin and other oral anticoagulants
May prolong prothrombin time; monitor INR.
Storage
Store dry powder below 30°C, protected from light and moisture. Store reconstituted suspension in a refrigerator (2-8°C). Do not freeze.
Overdose
Symptoms may include gastrointestinal distress (nausea, vomiting, diarrhea) and disturbances in fluid and electrolyte balance. Renal impairment and crystalluria may occur in cases of very high doses or pre-existing renal dysfunction. Treatment is symptomatic and supportive; hemodialysis can be used to remove amoxicillin from circulation.
Pregnancy & Lactation
Pregnancy Category B. Amoxicillin crosses the placenta but is not associated with an increased risk of birth defects. Use during pregnancy if clearly needed. Excreted in breast milk in small amounts; generally considered safe during lactation, but monitor infant for diarrhea, candidiasis, or allergic reactions.
