Description
Overview of the medicine
Aponia-1000 mg tablet contains Amoxicillin, a penicillin-type antibiotic used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
1000 mg every 8-12 hours, depending on the severity of infection.
Elderly
No specific dose adjustment for elderly patients with normal renal function; monitor renal function.
Renal_impairment
Dosage adjustment required based on creatinine clearance; e.g., CrCl 10-30 mL/min, 500 mg every 12 hours.
How to Take
Take orally with or without food. For best absorption, administer at the start of a meal.
Mechanism of Action
Amoxicillin works by inhibiting bacterial cell wall synthesis, leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Approximately 30 minutes to 1 hour.
Excretion
Mainly excreted unchanged by the kidneys via tubular secretion and glomerular filtration.
Half life
About 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly absorbed from the gastrointestinal tract, bioavailability of about 70-90%.
Metabolism
Limited hepatic metabolism; primarily forms inactive penicilloic acid.
Side Effects
Contraindications
- Hypersensitivity to amoxicillin or other penicillins
- History of severe hypersensitivity reaction to other beta-lactam antibiotics
Drug Interactions
Warfarin
May increase the anticoagulant effect of warfarin, requiring monitoring of INR.
Allopurinol
Increased risk of skin rash when co-administered with allopurinol.
Methotrexate
May reduce the renal clearance of methotrexate, potentially increasing its toxicity.
Oral Contraceptives
May reduce the efficacy of oral contraceptives.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms include gastrointestinal upset (nausea, vomiting, diarrhea), and in rare cases, crystalluria or renal impairment. Treatment is supportive and symptomatic; hemodialysis can be used to remove amoxicillin from circulation.
Pregnancy & Lactation
Generally considered safe for use during pregnancy (Category B). Small amounts are excreted in breast milk; generally considered compatible with breastfeeding, but monitor infant for potential adverse effects (e.g., diarrhea, candidiasis).
