Description
Overview of the medicine
Aponia 500 mg Tablet is a non-opioid analgesic and antipyretic medication used for symptomatic relief of mild to moderate pain and fever. It contains Aponia Hydrochloride as the active ingredient.
Uses & Indications
Dosage
Adults
500 mg to 1000 mg every 4-6 hours as needed. Maximum daily dose 4000 mg (4 tablets).
Elderly
Same as adults, but caution for renal impairment. Dose adjustment may be required.
Renal_impairment
Creatinine clearance 10-50 mL/min: dose interval of 6 hours. Creatinine clearance <10 mL/min: dose interval of 8 hours.
How to Take
Oral. Can be taken with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Aponia primarily acts by inhibiting prostaglandin synthesis in the central nervous system (CNS) and to a lesser extent, peripherally. It produces analgesia by elevating the pain threshold and antipyresis through its action on the hypothalamic heat-regulating center.

Excretion
Excreted predominantly in the urine as glucuronide and sulfate conjugates; less than 5% is excreted unchanged.
Half life
2-3 hours
Absorption
Rapidly and completely absorbed from the gastrointestinal tract, peak plasma concentrations reached within 30-60 minutes.
Metabolism
Primarily metabolized in the liver, mainly through glucuronidation and sulfation. A small portion is metabolized by cytochrome P450 to a reactive intermediate (NAPQI), which is detoxified by glutathione.

Contraindications
- Hypersensitivity to Aponia or any component of the formulation
- Severe hepatic impairment
Drug Interactions
Alcohol
Increases the risk of hepatotoxicity with chronic heavy alcohol use.
Warfarin
May enhance the anticoagulant effect of warfarin with prolonged use, increasing risk of bleeding.
Other hepatotoxic drugs
Increases the risk of liver damage.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.

Symptoms include nausea, vomiting, abdominal pain, and liver damage (jaundice, liver tenderness) in severe cases. Antidote is N-acetylcysteine. Seek immediate medical attention.
