Apuldon 5 mg Suspension contains Domperidone, which is used to treat nausea and vomiting, and to improve gastric motility in conditions like dyspepsia.
Uses & Indications
Dosage
Adults
Typically 10 ml (10 mg) 3 times daily, 15-30 minutes before meals. Maximum 30 mg/day.
Elderly
Same as adults, but caution advised due to potential for increased cardiac risk.
Renal_impairment
Dosage reduction may be necessary. For severe impairment (CrCl <30 ml/min), frequency should be reduced to 1-2 times daily depending on severity.
How to Take
Shake the bottle well before use. Take orally, preferably 15-30 minutes before meals. Do not exceed the prescribed dose.
Mechanism of Action
Domperidone is a dopamine D2 and D3 receptor antagonist. It blocks dopamine receptors in the chemoreceptor trigger zone (CTZ) and in the stomach, thereby exhibiting antiemetic and prokinetic effects.

- Patients with known prolonged cardiac conduction intervals (e.g., QTc), significant electrolyte disturbances, or underlying cardiac diseases (e.g., congestive heart failure)
- Concomitant use with strong CYP3A4 inhibitors or QTc-prolonging drugs
Drug Interactions
Antacids and H2-receptor antagonists
Reduce bioavailability of Domperidone if taken concomitantly. Administer Domperidone before meals and antacids/H2 blockers after meals.
Strong CYP3A4 inhibitors (e.g., ketoconazole, erythromycin)
Increased plasma levels of Domperidone, leading to increased risk of QTc prolongation.
QTc prolonging drugs (e.g., certain antiarrhythmics, antipsychotics)
Additive effect on QTc interval, increasing risk of arrhythmias.
