Description
Overview of the medicine
Aristo-D3 2000 IU Tablet contains Cholecalciferol, which is Vitamin D3. It is primarily used to treat and prevent vitamin D deficiency, promote calcium absorption, and support bone health. It plays a crucial role in maintaining strong bones and teeth, supporting immune function, and regulating cell growth.
Uses & Indications
Dosage
Adults
Typical daily dose for deficiency prevention is 2000 IU to 4000 IU. For treatment of deficiency, higher doses may be prescribed by a physician.
Elderly
Similar to adult dosage; higher doses may be needed due to reduced skin synthesis and absorption.
Renal_impairment
Use with caution in severe renal impairment; monitoring of calcium and phosphate levels is necessary. Active vitamin D metabolites may be preferred in severe cases.
How to Take
Swallow the tablet whole with water. Can be taken with or without food, but taking it with a meal containing fat may enhance absorption.
Mechanism of Action
Cholecalciferol (Vitamin D3) is a fat-soluble vitamin that helps regulate calcium and phosphate levels in the body. It converts into its active form, calcitriol, in the liver and kidneys. Calcitriol then acts on the intestines to increase calcium and phosphate absorption, on bones to regulate bone remodeling, and on kidneys to reduce calcium and phosphate excretion. It also has effects on immune cells and other tissues.
Pharmacokinetics
Onset
Therapeutic effects may take several days to weeks to manifest fully, particularly for bone health.
Excretion
Excreted primarily in the bile and feces, with a small amount excreted in urine.
Half life
The elimination half-life of Cholecalciferol itself is approximately 19-48 hours, but its active metabolite (calcitriol) has a half-life of 3-6 hours, and the storage form (25-hydroxyvitamin D) has a half-life of ~2-3 weeks.
Absorption
Readily absorbed from the small intestine in the presence of bile salts.
Metabolism
Metabolized in the liver to 25-hydroxyvitamin D (calcifediol) and then in the kidneys to 1,25-dihydroxyvitamin D (calcitriol, the active form).
Side Effects
Contraindications
- Hypercalcemia (high calcium levels).
- Vitamin D toxicity.
- Hypersensitivity to Cholecalciferol or any components of the formulation.
Drug Interactions
Corticosteroids
May decrease vitamin D effects.
Thiazide diuretics
May increase the risk of hypercalcemia.
Cardiac glycosides (e.g., digoxin)
Hypercalcemia can increase the risk of arrhythmias.
Bile acid sequestrants (e.g., cholestyramine)
May impair absorption of vitamin D.
Anticonvulsants (e.g., phenytoin, barbiturates)
May increase vitamin D metabolism, reducing its effectiveness.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms include hypercalcemia (nausea, vomiting, thirst, polyuria, constipation, muscle weakness, confusion, kidney stones). Treatment involves discontinuing vitamin D, low-calcium diet, hydration, and possibly corticosteroids or diuretics to manage calcium levels. Severe cases require hospitalization.
Pregnancy & Lactation
Generally considered safe and necessary during pregnancy and lactation within recommended doses to ensure adequate maternal and fetal/infant vitamin D levels. High doses should be avoided without medical supervision.

Clinical Trials
Numerous clinical trials support the efficacy of Cholecalciferol in preventing and treating vitamin D deficiency and its related bone health benefits. Specific trials for Aristo-D3 are typically bioequivalence studies.
Lab Monitoring
- Serum 25-hydroxyvitamin D levels (to assess vitamin D status).
- Serum calcium levels (especially during high-dose therapy).
- Serum phosphate levels.
- Kidney function tests (e.g., creatinine) in patients with renal impairment or long-term high-dose therapy.
Doctor Notes
- Emphasize the importance of adequate vitamin D levels for overall health, beyond just bone health.
- Advise patients on lifestyle modifications (diet, sun exposure) alongside supplementation.
- Caution patients about self-medicating with very high doses.
Patient Guidelines
- Take as directed by your doctor or pharmacist.
- Do not exceed the recommended dose without medical advice.
- Report any unusual symptoms, especially signs of high calcium, to your doctor.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it's close to the time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Aristo-D3 2000 IU Tablet is not known to affect the ability to drive or operate machinery.
Lifestyle Advice
- Ensure adequate sun exposure (if appropriate and safe) to naturally boost vitamin D levels.
- Consume a balanced diet rich in calcium and vitamin D (e.g., fatty fish, fortified dairy products).
- Regular weight-bearing exercise for bone health.
