Overview of the medicine
Anastrozole (Arotril) is a potent and selective non-steroidal aromatase inhibitor used in the treatment of hormone-receptor-positive breast cancer in postmenopausal women.
Uses & Indications
Dosage
Adults
1 mg orally once daily.
Elderly
No dosage adjustment is required for elderly patients.
Renal_impairment
No dosage adjustment is necessary for patients with mild to moderate renal impairment. Use with caution in patients with severe renal impairment (creatinine clearance <20 mL/min) due to limited data.
How to Take
Arotril tablets should be taken orally, with or without food. It is recommended to take the tablet at approximately the same time each day.
Mechanism of Action
Anastrozole selectively inhibits the aromatase enzyme, which is responsible for converting androgens to estrogens in peripheral tissues. By reducing estrogen levels, it slows the growth of hormone-dependent breast cancer.

Onset
Significant reduction in circulating estrogen levels within 24 hours.
Excretion
Approximately 10% excreted unchanged in urine, and 60% as metabolites within 72 hours.
Half life
Approximately 40-50 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract; peak plasma concentrations are usually reached within 2 hours.
Metabolism
Primarily metabolized in the liver via N-dealkylation, hydroxylation, and glucuronidation. Major metabolite is triazole, which is inactive.
Side Effects
Contraindications

- •Patients with known hypersensitivity to anastrozole or any excipients of the tablet.
Drug Interactions
Tamoxifen
Concomitant use with tamoxifen or estrogen-containing therapies should be avoided as they may reduce anastrozole's efficacy.
Estrogen-containing therapies
Should not be co-administered as they negate the pharmacological action of anastrozole.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
There is no specific antidote for anastrozole overdose. Treatment should be symptomatic and supportive, including general supportive measures and frequent monitoring of vital signs.
Pregnancy & Lactation
Arotril is contraindicated in pregnant or breastfeeding women. Women of childbearing potential should use effective contraception during treatment and for a period after.
