Description
Overview of the medicine
Asibalin 50 mg Capsule contains Pregabalin, an anticonvulsant and neuropathic pain agent. It is used to treat neuropathic pain, fibromyalgia, partial onset seizures, and generalized anxiety disorder.
Uses & Indications
Dosage
Adults
Initial dose is typically 75 mg twice daily or 50 mg three times daily (150 mg/day). May be titrated up to 300-600 mg/day, usually in 2-3 divided doses, based on individual response and tolerability.
Elderly
Reduced doses may be required due to potential age-related decrease in renal function. Dosage should be adjusted based on creatinine clearance.
Renal_impairment
Dosage must be adjusted based on creatinine clearance. For CrCl 30-60 mL/min, initial 75 mg/day in 2-3 divided doses; max 300 mg/day. For CrCl 15-30 mL/min, initial 25-50 mg/day in 1-2 divided doses; max 150 mg/day. For CrCl <15 mL/min, initial 25 mg once daily or 50 mg every other day; max 75 mg/day.
How to Take
Asibalin capsules should be swallowed whole with water, with or without food. Do not chew, crush, or open the capsule. It should be taken at regularly spaced intervals.
Mechanism of Action
Pregabalin binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, reducing the release of several excitatory neurotransmitters such as glutamate, norepinephrine, and substance P. This leads to a decrease in neuronal excitability and pain transmission.
Pharmacokinetics
Onset
Within a few days for neuropathic pain relief; may take longer for full anxiolytic or anticonvulsant effects.
Excretion
Primarily renal excretion as unchanged drug; less than 0.1% excreted in feces.
Half life
Approximately 6.3 hours. Eliminated primarily by renal excretion.
Absorption
Rapidly absorbed after oral administration; peak plasma concentrations reached within 1 hour. Bioavailability is ≥90% and is independent of dose.
Metabolism
Negligible metabolism in humans; approximately 98% of the dose is excreted unchanged in the urine.
Side Effects
Contraindications
- Hypersensitivity to Pregabalin or any component of the formulation.
- Severe allergic reactions to other GABA analogues.
Drug Interactions
ACE Inhibitors (e.g., lisinopril, enalapril)
May increase the risk of angioedema when co-administered with pregabalin.
CNS Depressants (e.g., opioids, benzodiazepines, alcohol)
Concurrent use may enhance the depressant effects on the central nervous system, leading to increased sedation, dizziness, and respiratory depression.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and coma. Treatment is symptomatic and supportive. Hemodialysis may be beneficial in severe cases.
Pregnancy & Lactation
Pregnancy Category C. Should only be used during pregnancy if the potential benefit justifies the potential risk to the fetus. Pregabalin is excreted in human breast milk; therefore, breastfeeding is not recommended during treatment.
