Description
Overview of the medicine
Asibalin 75 mg Capsule contains Pregabalin, an anticonvulsant and anxiolytic drug used to treat neuropathic pain, fibromyalgia, partial-onset seizures, and generalized anxiety disorder.
Uses & Indications
Dosage
Adults
Initial dose 75 mg twice daily or 50 mg three times daily (150 mg/day). May be increased to 150 mg twice daily or 100 mg three times daily (300 mg/day) within 1 week. Max dose 300 mg twice daily or 200 mg three times daily (600 mg/day).
Elderly
Dosage adjustments may be necessary due to potential age-related decrease in renal function. Start with a lower dose.
Renal_impairment
Dosage must be adjusted based on creatinine clearance. For CrCl 30-60 mL/min, 75 mg once or twice daily. For CrCl 15-30 mL/min, 25-50 mg once daily. For CrCl <15 mL/min, 25 mg once daily or every other day.
How to Take
Asibalin capsules should be swallowed whole with water, with or without food. Do not chew, crush, or open the capsule.
Mechanism of Action
Pregabalin binds to the alpha-2-delta subunit of voltage-gated calcium channels in the central nervous system, reducing the release of several excitatory neurotransmitters like glutamate, noradrenaline, and substance P. This results in reduced neuronal excitability, leading to its analgesic, anxiolytic, and anticonvulsant effects.
Pharmacokinetics
Onset
Pain relief can begin within 1 week for neuropathic pain. Anxiolytic effects may be seen earlier.
Excretion
Excreted almost entirely in the urine as unchanged drug. Renal elimination is the major route.
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed after oral administration; peak plasma concentrations reached within 1 hour. Bioavailability is ≥ 90% and is independent of dose.
Metabolism
Minimally metabolized in humans (<2% of dose). Primarily excreted as unchanged drug.
Side Effects
Contraindications
- Hypersensitivity to pregabalin or any component of the formulation.
- Severe kidney impairment without dose adjustment.
Drug Interactions
CNS depressants (e.g., alcohol, benzodiazepines, opioids)
Increased risk of sedation and respiratory depression.
Thiazolidinedione antidiabetics (e.g., pioglitazone, rosiglitazone)
May increase risk of weight gain and peripheral edema.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and coma. Treatment is supportive and symptomatic; hemodialysis may be beneficial in some cases.
Pregnancy & Lactation
