Description
Overview of the medicine
Azalid 200 mg Suspension contains Azithromycin, a macrolide antibiotic used to treat a wide range of bacterial infections including those of the respiratory tract, skin, and sexually transmitted infections.
Uses & Indications
Dosage
Adults
For adults, a typical course for infections is 500 mg (12.5 ml of 200 mg/5 ml suspension) as a single dose on Day 1, followed by 250 mg (6.25 ml) once daily on Days 2-5, or 500 mg once daily for 3 days. Doses are typically adjusted for children based on weight.
Elderly
No specific dosage adjustment is recommended for elderly patients with normal renal and hepatic function.
Renal_impairment
No dosage adjustment is necessary for patients with mild to moderate renal impairment (creatinine clearance >10-80 mL/min). Caution is advised in severe renal impairment.
How to Take
Administer orally, with or without food. Shake the bottle well before each use. Use the provided measuring device for accurate dosing. Finish the entire prescribed course.
Mechanism of Action
Azithromycin inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit of susceptible microorganisms, thereby interfering with the translocation of peptides.
Pharmacokinetics
Onset
Clinical improvement usually observed within 2-3 days of initiation.
Excretion
Mainly excreted unchanged in the bile (approximately 50%), with a small amount excreted via the kidneys (about 6%).
Half life
The terminal elimination half-life is 2-4 days, allowing for once-daily dosing.
Absorption
Rapidly absorbed from the gastrointestinal tract; bioavailability is about 37%. Peak plasma concentrations are reached in 2-3 hours.
Metabolism
Primarily metabolized in the liver by demethylation into inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to azithromycin, erythromycin, any macrolide or ketolide antibiotic, or to any component of the formulation.
- •History of cholestatic jaundice/hepatic dysfunction associated with prior azithromycin therapy.
Drug Interactions
Warfarin
Increased anticoagulant effect. Monitor Prothrombin Time (PT)/INR closely.
Nelfinavir
Increased azithromycin serum concentrations. No dosage adjustment required.
Antacids (containing aluminum or magnesium)
Decreased peak plasma levels of azithromycin. Administer azithromycin at least 2 hours before or 4 hours after antacids.
QT-prolonging drugs (e.g., amiodarone, quinidine)
Potential for additive QT interval prolongation and increased risk of cardiac arrhythmias.
Storage
Store the dry powder below 30°C, protect from light and moisture. After reconstitution, store the suspension in a refrigerator (2-8°C) and discard any unused portion after the specified period (e.g., 5-10 days). Do not freeze.
Overdose
Symptoms of overdose may include severe nausea, vomiting, diarrhea, and reversible hearing loss. Management involves gastric lavage, symptomatic and supportive measures.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed and the potential benefits outweigh the risks. Azithromycin is excreted in human breast milk; caution should be exercised when administered to a nursing woman.
