Blast-125mg Oral Solution is a broad-spectrum antibiotic used to treat a variety of bacterial infections in children.
Uses & Indications
Dosage
Adults
Dosage for specific infections varies; generally 250-500 mg every 8 hours. 125 mg solution is primarily for pediatric use.
Elderly
Dosage adjustment may be required based on renal function; consult physician.
Renal_impairment
Severe renal impairment (CrCl < 30 mL/min): Dose reduction or increased dosing interval is necessary.
How to Take
Shake well before use. Administer orally with or without food. Use the provided measuring device for accurate dosing.
Mechanism of Action
It works by inhibiting the synthesis of the bacterial cell wall, leading to bacterial lysis and death.
Pharmacokinetics
Onset
Onset of action within 30-60 minutes.
Excretion
Primarily excreted unchanged in the urine, with some biliary excretion.
Half life
Elimination half-life is approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly absorbed after oral administration, reaching peak plasma concentrations within 1-2 hours.
Metabolism
Partially metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to penicillins or any component of the formulation.
- History of severe hypersensitivity reactions to other beta-lactam antibiotics (e.g., cephalosporins, carbapenems).
Drug Interactions
Probenecid
Concomitant use with probenecid decreases renal tubular secretion of blast-125mg, leading to increased and prolonged plasma concentrations.
Methotrexate
May reduce the renal clearance of methotrexate, potentially increasing toxicity.
Oral anticoagulants
May potentiate the anticoagulant effect, requiring monitoring of INR/prothrombin time.
Storage
Store the unmixed powder below 30°C in a dry place, protected from light and moisture. Store the reconstituted solution in a refrigerator (2-8°C). Keep out of reach of children.
Overdose
Symptoms of overdose include gastrointestinal effects (nausea, vomiting, diarrhea) and disturbances of fluid and electrolyte balance. In isolated cases, renal failure and convulsions have been reported. Management involves gastric lavage, activated charcoal, and symptomatic and supportive treatment. Hemodialysis may be used to remove the drug from circulation.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus. However, use in pregnancy should only be considered if clearly needed. Excreted in breast milk; caution is advised when administered to a nursing mother.
