Description
Overview of the medicine
Ceclav-500 mg tablet is a broad-spectrum antibiotic containing Amoxicillin and Clavulanate Potassium. It is used to treat various bacterial infections, including respiratory tract infections, urinary tract infections, skin and soft tissue infections, and otitis media.
Uses & Indications
Dosage
Adults
One 500 mg/125 mg tablet every 8-12 hours, or as directed by physician. Duration usually 5-14 days depending on infection severity.
Elderly
Dosage adjustment usually not required unless severe renal impairment is present.
Renal_impairment
Dosage adjustment needed based on creatinine clearance. e.g., CrCl 10-30 mL/min: 500 mg/125 mg every 12 hours; CrCl <10 mL/min: 500 mg/125 mg every 24 hours.
How to Take
Take the tablet orally with a glass of water, preferably at the start of a meal to minimize potential gastrointestinal intolerance. Do not crush or chew.
Mechanism of Action
Amoxicillin is a beta-lactam antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs). Clavulanate Potassium is a beta-lactamase inhibitor that protects amoxicillin from degradation by bacterial beta-lactamase enzymes, thus extending its antibacterial spectrum.

Onset
Approximately 1 hour (antibacterial activity)
Excretion
Primarily excreted unchanged in the urine (amoxicillin) and through urine (clavulanate metabolites and unchanged drug).
Half life
Amoxicillin: 1-1.3 hours; Clavulanate: 1 hour
Absorption
Both amoxicillin and clavulanate potassium are well absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are achieved in approximately 1-2 hours.
Metabolism
Amoxicillin is minimally metabolized; Clavulanate is extensively metabolized.
Side Effects
Contraindications
- •Hypersensitivity to penicillin or other beta-lactam antibiotics
- •History of cholestatic jaundice/hepatic dysfunction associated with Amoxicillin/Clavulanate Potassium
- •Infectious mononucleosis (may cause a rash)
Drug Interactions
Probenecid
Decreases renal tubular secretion of amoxicillin, leading to increased and prolonged blood levels of amoxicillin. Concomitant use is not recommended.
Allopurinol
Increased incidence of rash in patients receiving allopurinol and amoxicillin concurrently.
Methotrexate
May decrease methotrexate excretion, potentially increasing toxicity. Monitor methotrexate levels.
Oral Contraceptives
May reduce the efficacy of oral contraceptives; advise patients to use an additional non-hormonal method of contraception.
Anticoagulants (e.g., Warfarin)
May prolong prothrombin time (INR). Close monitoring of INR is advised.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include gastrointestinal symptoms (nausea, vomiting, diarrhea) and disturbances of the fluid and electrolyte balance. Renal impairment may lead to convulsions. Treatment is supportive; hemodialysis can remove amoxicillin and clavulanate from circulation.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown harm to the fetus, but there are no adequate and well-controlled studies in pregnant women. Use only if clearly needed. Amoxicillin and clavulanate are excreted into breast milk in small amounts; generally considered safe during lactation, but monitor for infant side effects like diarrhea, candidiasis.
