Description
Overview of the medicine
Ceftazidime is a third-generation cephalosporin antibiotic used to treat various bacterial infections, especially Gram-negative infections, including pseudomonal infections.
Uses & Indications
Dosage
Adults
1-2 grams IV or IM every 8-12 hours; for severe infections, up to 6 grams daily in divided doses.
Elderly
Dosage adjustment may be required based on renal function and overall health status.
Renal_impairment
Dosage must be reduced based on creatinine clearance. Consult a dosage chart for specific recommendations.
How to Take
Administered by slow intravenous (IV) injection (3-5 minutes), intermittent IV infusion (20-30 minutes), or deep intramuscular (IM) injection.
Mechanism of Action
Ceftazidime inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bactericidal action against susceptible organisms.
Pharmacokinetics
Onset
Rapid, typically within minutes of IV administration.
Excretion
Mainly via the kidneys, almost entirely by glomerular filtration.
Half life
Approximately 1.5 to 2 hours in adults with normal renal function.
Absorption
Rapid and complete absorption after intramuscular (IM) or intravenous (IV) administration.
Metabolism
Minimally metabolized; primarily excreted unchanged.
Side Effects
Contraindications
- •Hypersensitivity to ceftazidime, any other cephalosporin, or any excipients in the formulation.
- •History of severe hypersensitivity reaction to any other type of beta-lactam antibacterial agent (e.g., penicillins, carbapenems).
Drug Interactions
Aminoglycosides
Increased risk of nephrotoxicity, especially with high doses or pre-existing renal impairment.
Chloramphenicol
Antagonistic effect observed in vitro; avoid concomitant use.
Oral anticoagulants
Enhanced anticoagulant effect may occur, requiring close monitoring of INR/PT.
Loop diuretics (e.g., Furosemide)
May increase nephrotoxicity when co-administered with aminoglycosides.
Storage
Store unconstituted powder below 25°C (77°F) and protect from light. Reconstituted solutions should be used immediately or stored as per manufacturer's guidelines (e.g., 18-24 hours at room temperature or 7 days refrigerated).
Overdose
Symptoms of overdose may include neurological sequelae such as encephalopathy, convulsions, and coma, especially in patients with renal impairment. Treatment is supportive; hemodialysis may be beneficial in removing the drug from circulation.
Pregnancy & Lactation
Use during pregnancy only if clearly indicated and the potential benefits outweigh the risks. Excreted in breast milk in small amounts; use with caution during lactation.
