Description
Overview of the medicine
Cenolon 55 mcg Nasal Spray contains Fluticasone Furoate, a synthetic trifluorinated corticosteroid, indicated for the treatment of seasonal and perennial allergic rhinitis symptoms in adults and children.
Uses & Indications
Dosage
Adults
Initially, 2 sprays in each nostril once daily. May be reduced to 1 spray in each nostril once daily for maintenance.
Elderly
No dosage adjustment required.
Renal_impairment
No dosage adjustment required.
How to Take
For intranasal use only. Shake gently before use. Prime the spray pump before first use or if not used for more than 30 days. Spray into each nostril while blocking the other, avoiding contact with eyes.
Mechanism of Action
Fluticasone Furoate is a synthetic trifluorinated corticosteroid with potent anti-inflammatory activity. It binds to glucocorticoid receptors, leading to inhibition of multiple cell types (e.g., mast cells, eosinophils, neutrophils, macrophages, lymphocytes) and mediators (e.g., histamine, eicosanoids, leukotrienes, cytokines) involved in allergic and inflammatory responses.
Pharmacokinetics
Onset
Symptomatic relief within 8-24 hours; full effect may take several days of continuous use.
Excretion
Primarily excreted in feces, with a small amount in urine.
Half life
Approximately 15.1 hours (systemic half-life).
Absorption
Low systemic absorption (typically <0.5%) after nasal administration due to extensive first-pass metabolism.
Metabolism
Extensively metabolized by CYP3A4 in the liver via hydrolysis.
Side Effects
Contraindications
- •Hypersensitivity to fluticasone furoate or any components of the spray.
Drug Interactions
Ritonavir
Concurrent use with strong CYP3A4 inhibitors like ritonavir may increase systemic exposure to fluticasone furoate, leading to potential systemic corticosteroid effects. Concomitant use should be avoided unless the benefit outweighs the increased risk of systemic corticosteroid side effects.
Ketoconazole
Strong CYP3A4 inhibitors (e.g., ketoconazole) can increase systemic exposure to fluticasone furoate. Use with caution.
Storage
Store at room temperature (20°C to 25°C), away from moisture and direct heat. Do not freeze.
Overdose
Acute overdose is unlikely due to the low systemic absorption and topical route of administration. Chronic overdose may lead to signs of hypercorticism. Treatment is symptomatic and supportive; gradual withdrawal if chronic overdose occurs.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown whether fluticasone furoate is excreted in human milk; use with caution in nursing mothers.
