Description
Overview of the medicine
Clavucure 125 mg Suspension is an antibiotic medication combining amoxicillin and clavulanic acid, used to treat a wide range of bacterial infections including respiratory tract infections, urinary tract infections, skin and soft tissue infections, and ear infections. Clavulanic acid helps amoxicillin to work effectively against bacteria that produce beta-lactamase enzymes.
Uses & Indications
Dosage
Adults
This strength (125mg Amoxicillin + 31.25mg Clavulanic Acid per 5ml) is primarily for pediatric use. Adults typically require higher strengths.
Elderly
This strength is not typically prescribed for elderly patients. Dosage for elderly patients using other strengths should be adjusted based on renal function.
Renal_impairment
For patients with renal impairment, regardless of age, dosage adjustments are crucial. Consult a physician for specific recommendations.
How to Take
Shake the bottle well before each use. Administer orally using a measuring spoon or oral syringe. Can be taken with or without food; taking with food may reduce gastrointestinal discomfort.
Mechanism of Action
Amoxicillin is a beta-lactam antibiotic that inhibits bacterial cell wall synthesis. Clavulanic acid is a beta-lactamase inhibitor that protects amoxicillin from degradation by beta-lactamase enzymes produced by resistant bacteria, thereby extending amoxicillin's spectrum of activity.
Pharmacokinetics
Onset
Onset of action typically occurs within 1-2 hours.
Excretion
Primarily excreted unchanged by the kidneys (amoxicillin) and renally as metabolites (clavulanic acid).
Half life
Amoxicillin: 1-1.5 hours; Clavulanic acid: 1 hour.
Absorption
Both amoxicillin and clavulanic acid are well absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are achieved within approximately 1-2 hours.
Metabolism
Amoxicillin is minimally metabolized. Clavulanic acid is extensively metabolized.
Side Effects
Contraindications
- Known hypersensitivity to penicillin or cephalosporin antibiotics or to any component of the formulation
- History of cholestatic jaundice/hepatic dysfunction associated with amoxicillin/clavulanic acid therapy
Drug Interactions
Probenecid
Increases and prolongs amoxicillin blood levels.
Allopurinol
May increase the incidence of skin rash.
Methotrexate
May increase methotrexate toxicity due to reduced renal clearance.
Oral contraceptives
May reduce the efficacy of oral contraceptives (advise additional contraception).
Anticoagulants (e.g., Warfarin)
May prolong prothrombin time; monitor INR closely.
Storage
Store dry powder below 25°C in a cool, dry place, away from direct sunlight. Once reconstituted, store in a refrigerator (2-8°C) and discard any unused portion after 7-10 days. Do not freeze.
Overdose
Symptoms may include gastrointestinal upset (nausea, vomiting, diarrhea), and disturbances of fluid and electrolyte balance. Treatment is symptomatic and supportive. Hemodialysis can be used to remove amoxicillin/clavulanic acid from circulation.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed. Excreted in breast milk, use with caution during lactation.
