Clavucure-500-mg-tablet is a broad-spectrum antibiotic combination of amoxicillin and clavulanic acid, used to treat various bacterial infections, including those caused by beta-lactamase producing bacteria.
Uses & Indications
Dosage
Adults
The usual adult dose is 500 mg amoxicillin/125 mg clavulanic acid every 8 hours, or 875 mg amoxicillin/125 mg clavulanic acid every 12 hours, depending on the severity of the infection.
Elderly
No specific dose adjustment is required in elderly patients with normal renal function. Dose should be adjusted in case of renal impairment.
Renal_impairment
Dose adjustment is necessary based on creatinine clearance. For CrCl 10-30 mL/min, 500 mg/125 mg every 12 hours. For CrCl <10 mL/min, 500 mg/125 mg every 24 hours.
How to Take
Take Clavucure-500-mg-tablet orally at the start of a meal to minimize potential gastrointestinal intolerance and to optimize absorption.
Mechanism of Action
Amoxicillin inhibits bacterial cell wall synthesis, leading to cell lysis and death. Clavulanic acid inhibits beta-lactamase enzymes, which are produced by some bacteria to break down amoxicillin, thus extending amoxicillin's antibacterial spectrum.
Pharmacokinetics
Onset
Peak plasma concentrations are usually achieved within 1 to 2 hours after oral administration.
Excretion
Both components are primarily excreted by the kidneys, with amoxicillin largely excreted unchanged.
Half life
The elimination half-life is approximately 1 to 1.5 hours for amoxicillin and about 1 hour for clavulanic acid.
Absorption
Both amoxicillin and clavulanic acid are well absorbed from the gastrointestinal tract after oral administration. Absorption is optimized when taken at the start of a meal.
Metabolism
Amoxicillin is partially metabolized, while clavulanic acid is extensively metabolized.

Contraindications
- History of hypersensitivity to amoxicillin, clavulanic acid, or any other penicillin antibiotic
- History of severe hypersensitivity reactions (e.g., anaphylaxis) to any beta-lactam antibiotic (e.g., cephalosporins)
- History of cholestatic jaundice or hepatic dysfunction associated with amoxicillin/clavulanic acid therapy
Drug Interactions
Allopurinol
Increased incidence of rash with concomitant use.
Methotrexate
May increase methotrexate toxicity; monitor for signs of toxicity.
Oral contraceptives
May reduce the efficacy of oral contraceptives; advise additional contraception.
Warfarin and other oral anticoagulants
May prolong prothrombin time; monitor INR closely.

Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.

Symptoms of overdose include gastrointestinal effects (e.g., abdominal pain, vomiting, diarrhea) and fluid and electrolyte imbalance. Rarely, renal impairment or convulsions may occur. Treatment is symptomatic and supportive, and hemodialysis may be used to remove the drug from circulation.

Pregnancy Category B. While studies have not shown harm, use during pregnancy should only be if clearly needed. Both amoxicillin and clavulanic acid are excreted into breast milk; use with caution in nursing mothers.
