Overview of the medicine
Comfy 1 mg Tablet is an anxiolytic medicine containing Lorazepam, used to treat anxiety disorders, insomnia, and for pre-operative sedation. It works by enhancing the activity of a certain natural chemical (GABA) in the body, which helps to calm the brain and nerves.
Uses & Indications
Dosage
Adults
Anxiety: 1-4 mg daily in divided doses. Insomnia: 1-2 mg at bedtime. Pre-operative: 2-4 mg the night before or 1-2 hours before surgery.
Elderly
Initially, a lower dose of 0.5-1 mg daily in divided doses, adjusted as needed and tolerated.
Renal_impairment
No specific dose adjustment required, but caution is advised. Initiate with lower doses.
How to Take
Administer orally with or without food. For insomnia, take at bedtime. Do not crush or chew the tablet unless advised by a doctor.
Mechanism of Action
Lorazepam potentiates the effect of GABA (gamma-aminobutyric acid), an inhibitory neurotransmitter, at the GABA-A receptor, leading to increased chloride ion influx, hyperpolarization of the neuron, and decreased neuronal excitability.

Oral: 20-30 minutes; IV: 1-5 minutes.
Excretion
Excreted renally as glucuronide conjugates.
Half life
Approximately 10-20 hours.
Absorption
Readily absorbed after oral administration; peak plasma concentrations achieved in 1-2 hours.
Metabolism
Primarily hepatic conjugation with glucuronic acid; no active metabolites.
Side Effects
Contraindications
- •Known hypersensitivity to benzodiazepines or any component of the formulation

Drug Interactions
Opioids
Increased risk of sedation, respiratory depression, coma, and death.
Valproate
Increases lorazepam plasma levels.
Probenecid
Increases lorazepam plasma levels and prolongs its half-life.
Alcohol and other CNS depressants
Additive CNS depressant effects.
Storage
Store at controlled room temperature (20-25°C), protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms include somnolence, confusion, diminished reflexes, coma, and cardiorespiratory depression. Management includes supportive care, monitoring vital signs, and if severe, administration of flumazenil, a benzodiazepine receptor antagonist.
Pregnancy & Lactation
Pregnancy Category D. Lorazepam can cause fetal harm. Avoid during pregnancy. Excreted in breast milk, avoid during lactation.
