D-Revive 40000 IU Capsule contains Cholecalciferol (Vitamin D3), a fat-soluble vitamin essential for calcium and phosphate homeostasis and bone health. It is primarily used for the treatment and prevention of Vitamin D deficiency.
Uses & Indications
Dosage
Adults
For severe deficiency: 40000 IU once weekly for 6-8 weeks, followed by a maintenance dose as determined by a physician. For maintenance: 20000 IU every two weeks or 40000 IU once a month, depending on patient needs and serum levels.
Elderly
Similar to adult dosage; however, renal function and calcium levels should be monitored closely.
Renal_impairment
Use with caution. Dosage adjustment may be necessary based on serum calcium and phosphate levels. Active vitamin D metabolites may be preferred in severe renal failure.
How to Take
The capsule should be swallowed whole with water. It is best taken with the largest meal of the day to enhance absorption.
Mechanism of Action
Cholecalciferol (Vitamin D3) is metabolized in the liver to 25-hydroxycholecalciferol (calcidiol) and then in the kidneys to 1,25-dihydroxycholecalciferol (calcitriol), the active form. Calcitriol regulates serum calcium and phosphate levels by promoting absorption from the gut, reabsorption in the kidneys, and influencing bone mineralization.

Drug Interactions
Corticosteroids
May reduce Vitamin D action.
Cardiac glycosides
Increased risk of arrhythmia if hypercalcemia develops.
Thiazide diuretics
May increase the risk of hypercalcemia.
Orlistat, Cholestyramine, Mineral oil
May reduce absorption of Vitamin D and other fat-soluble vitamins.
Anticonvulsants (e.g., phenytoin, barbiturates)
May increase Vitamin D metabolism, reducing its effectiveness.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms include hypercalcemia (nausea, vomiting, loss of appetite, constipation, weakness, polyuria, polydipsia, headache). Severe cases can lead to renal impairment, cardiac arrhythmias, and soft tissue calcification. Treatment involves discontinuing Vitamin D, reducing calcium intake, ensuring adequate hydration, and in severe cases, corticosteroids or calcitonin may be used.

Consult a doctor before use. Generally considered safe in recommended doses during pregnancy and lactation to prevent deficiency, but excessive doses should be avoided due to potential risks of hypercalcemia to the fetus/infant.
