Dapclin is a brand name for Clindamycin, a lincosamide antibiotic used to treat a wide variety of bacterial infections.
Uses & Indications
Dosage
Adults
Oral: 150-450 mg every 6-8 hours. Max 1.8 g/day.
Elderly
No specific dose adjustment is generally required based on age alone, but renal and hepatic function should be monitored.
Renal_impairment
No dosage adjustment needed for mild to moderate renal impairment. For severe impairment, consider lengthening dosing interval.
How to Take
Oral capsules should be taken with a full glass of water to prevent esophageal irritation. Can be taken with or without food.
Mechanism of Action
Clindamycin binds to the 50S ribosomal subunit of susceptible bacteria, thereby inhibiting protein synthesis.
Pharmacokinetics
Onset
Varies by route of administration. Peak plasma concentrations are usually reached within 45-60 minutes after oral administration.
Excretion
Approximately 10-20% excreted in urine and 4-10% in feces as active drug and metabolites. Most is eliminated via bile and feces.
Half life
Approximately 2-3 hours in adults with normal renal and hepatic function.
Absorption
Rapidly and almost completely absorbed from the gastrointestinal tract (about 90% bioavailability for oral forms). Absorption is not significantly affected by food.
Metabolism
Primarily metabolized in the liver to active and inactive metabolites (e.g., N-demethylclindamycin and clindamycin sulfoxide).
Side Effects
Contraindications
- •History of hypersensitivity to clindamycin or lincomycin
- •History of antibiotic-associated colitis (especially pseudomembranous colitis)
Drug Interactions
Erythromycin
Antagonistic effect; do not use concurrently.
Kaolin-pectin antidiarrheals
May delay or decrease absorption of clindamycin.
Neuromuscular blocking agents
May enhance the neuromuscular blocking action, leading to increased respiratory depression.
Storage
Store in a cool, dry place, away from light. Do not store above 30°C.
Overdose
Symptoms of overdose include gastrointestinal disturbances. Treatment is symptomatic and supportive; hemodialysis is not effective.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed during pregnancy. Clindamycin is excreted in breast milk; caution should be exercised.
