Description
Overview of the medicine
Desloratadine (Desatrol) is a non-sedating antihistamine used for the relief of symptoms associated with allergic rhinitis and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
5 mg once daily.
Elderly
No specific dose adjustment is generally required, but caution is advised in elderly patients with renal or hepatic impairment.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), a reduced frequency of 5 mg every other day is recommended.
How to Take
Desatrol can be taken orally with or without food. Tablets should be swallowed whole with water.
Mechanism of Action
Desloratadine is a potent, long-acting, selective peripheral H1-receptor antagonist. It inhibits the release of histamine from mast cells and basophils and blocks the effects of histamine at the H1-receptor, thereby relieving allergy symptoms.

Onset
Onset of action within 1 hour.
Excretion
Eliminated equally in the urine and feces.
Half life
Approximately 27 hours.
Absorption
Rapidly and well absorbed after oral administration. Peak plasma concentrations are reached approximately 3 hours after administration.
Metabolism
Extensively metabolized to 3-hydroxydesloratadine, an active metabolite, primarily by CYP2C8 and CYP2C9.
Side Effects
Contraindications
- Hypersensitivity to the active substance desloratadine, to loratadine, or to any of the excipients.
- Patients with severe renal impairment (for syrup, consult physician).
Drug Interactions
Alcohol
Desloratadine does not potentiate the effects of alcohol.
Erythromycin
Coadministration with erythromycin may slightly increase desloratadine plasma levels, but this is not considered clinically significant.
Ketoconazole
Coadministration with ketoconazole may slightly increase desloratadine plasma levels, but this is not considered clinically significant.
Fluoxetine, Cimetidine
No clinically significant interactions have been observed with fluoxetine or cimetidine.

Store in a cool and dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.

In the event of an overdose, symptomatic and supportive treatment should be initiated. Desloratadine is not eliminated by hemodialysis.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Not recommended for use during lactation as desloratadine is excreted in breast milk.
