Emoxil 125 mg Pediatric Drop contains Amoxicillin, a penicillin antibiotic used to treat a wide range of bacterial infections in infants and children.
Uses & Indications
Dosage
Adults
Not directly applicable for 'pediatric drop' form. For pediatric patients (infants 3 months to children <40 kg): 20-50 mg/kg/day in 2-3 divided doses. For severe infections, up to 60 mg/kg/day may be given. Specific dose and duration should be determined by a physician. Example: 1.25 ml (125 mg) 2-3 times daily.
Elderly
Not applicable for this pediatric formulation.
Renal_impairment
Dosage adjustment is necessary for patients with severe renal impairment (creatinine clearance < 30 ml/min). Consult a physician for specific recommendations.
How to Take
Oral administration. Shake the bottle well before each use. Use the provided calibrated dropper to measure the exact prescribed dose. Can be taken with or without food.
Mechanism of Action
Amoxicillin acts by inhibiting the biosynthesis of the bacterial cell wall. It binds to penicillin-binding proteins (PBPs), leading to the disruption of bacterial cell wall formation and subsequent cell lysis and death.
Pharmacokinetics
Onset
Clinical effects usually seen within 30-60 minutes after administration.
Excretion
Mainly excreted unchanged in the urine via renal tubular secretion and glomerular filtration. A small amount is excreted in feces.
Half life
Approximately 1 to 1.5 hours in adults with normal renal function; longer in neonates and those with renal impairment.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, especially when taken orally. Peak plasma concentrations are achieved within 1-2 hours.
Metabolism
Approximately 10-25% of the dose is metabolized to inactive metabolites, primarily in the liver.
Side Effects
Contraindications
- •Hypersensitivity to amoxicillin, any other penicillin, or to any of the excipients.
- •History of severe immediate hypersensitivity reaction (e.g., anaphylaxis) to another beta-lactam agent (e.g., a cephalosporin, carbapenem or monobactam).
Drug Interactions
Probenecid
Concomitant use with probenecid is not recommended as it decreases the renal tubular secretion of amoxicillin, leading to increased and prolonged blood levels of amoxicillin.
Allopurinol
Concomitant administration of allopurinol during treatment with amoxicillin may increase the likelihood of allergic skin reactions.
Methotrexate
Penicillins may reduce the excretion of methotrexate, potentially increasing toxicity.
Tetracyclines
Tetracyclines and other bacteriostatic drugs may interfere with the bactericidal effects of penicillin and should be avoided.
Oral Anticoagulants
Prolongation of prothrombin time (INR) has been reported in patients receiving amoxicillin and oral anticoagulants. Careful monitoring is necessary.
Storage
Store the dry powder below 30°C, protected from light and moisture. After reconstitution, store the suspension in a refrigerator (2-8°C). Do not freeze. Keep out of reach of children.
Overdose
In case of overdose, gastrointestinal symptoms (nausea, vomiting, diarrhea) and disturbances of fluid and electrolyte balance may occur. Management involves gastric lavage, activated charcoal, and symptomatic and supportive treatment. Haemodialysis can be used to remove amoxicillin from circulation.
Pregnancy & Lactation
Pregnancy Category B. Amoxicillin can be used during pregnancy if clearly needed. Small amounts are excreted in breast milk; generally considered compatible with breastfeeding, but caution is advised. Consult a doctor.
