Overview of the medicine
Etodolac (Etoben) is a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, swelling, and stiffness caused by conditions such as osteoarthritis and rheumatoid arthritis. It works by blocking the production of certain natural substances in the body that cause inflammation.
Uses & Indications
Dosage
Adults
Acute pain: 200-400 mg every 6-8 hours. Osteoarthritis/Rheumatoid Arthritis: 300-500 mg 2-3 times daily or 400-600 mg twice daily. Max 1200 mg/day.
Elderly
Initiate at lower doses, monitor for adverse effects.
Renal_impairment
Use with caution; consider dose reduction in severe impairment. Not recommended for severe renal failure.
How to Take
Take orally with food or milk to reduce gastrointestinal upset.
Mechanism of Action
Etodolac inhibits cyclooxygenase (COX) enzymes, particularly showing preferential inhibition of COX-2, thereby reducing the synthesis of prostaglandins. Prostaglandins are mediators of inflammation, pain, and fever.

Excreted primarily via urine (approximately 60%) and feces (approximately 30-45%) as metabolites.

Side Effects
Contraindications
- •Hypersensitivity to etodolac or other NSAIDs
- •History of aspirin-induced asthma, urticaria, or allergic-type reactions
- •Active gastrointestinal bleeding or peptic ulcer disease
- •Severe renal or hepatic impairment
- •Third trimester of pregnancy
- •Coronary Artery Bypass Graft (CABG) surgery (peri-operative pain)
Drug Interactions
Lithium
Increased lithium plasma levels.
Cyclosporine
Increased risk of nephrotoxicity.
Methotrexate
Increased methotrexate toxicity.
ACE inhibitors, Diuretics
Reduced antihypertensive effect; increased risk of renal impairment.
Warfarin, other anticoagulants
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include lethargy, drowsiness, nausea, vomiting, epigastric pain, and gastrointestinal bleeding. Management involves symptomatic and supportive care. Gastric lavage or activated charcoal may be considered within 1 hour of ingestion.
Pregnancy & Lactation
Pregnancy: Avoid in the third trimester due to risk of premature closure of the ductus arteriosus. Use with caution in the first and second trimesters only if potential benefit outweighs risk. Lactation: Small amounts may pass into breast milk; generally not recommended.
