Description
Overview of the medicine
Etofax 60 mg Tablet contains Etoricoxib, a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain and inflammation in conditions like osteoarthritis, rheumatoid arthritis, and acute gout.
Uses & Indications
Dosage
Adults
Osteoarthritis: 30-60 mg once daily. Rheumatoid Arthritis/Ankylosing Spondylitis: 90 mg once daily. Acute Gouty Arthritis: 120 mg once daily (maximum 8 days). Acute Pain: 60-120 mg once daily, not exceeding 8 days for 120 mg dose.
Elderly
No specific dose adjustment required based on age, but caution is advised due to potential age-related decrease in renal function.
Renal_impairment
Moderate renal impairment (CrCl 30-80 mL/min) requires caution. Contraindicated in severe renal impairment (CrCl <30 mL/min).
How to Take
Administer orally, with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Etoricoxib selectively inhibits cyclooxygenase-2 (COX-2) enzyme, which is responsible for the production of prostaglandins involved in inflammation and pain. By inhibiting COX-2, it reduces pain and inflammation.

Onset
Approximately 1 hour for pain relief.
Excretion
Excreted primarily through the kidneys (major route) and in feces (minor route).
Half life
Approximately 22 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract. Peak plasma concentrations reached within approximately 1 hour.
Metabolism
Extensively metabolized in the liver, primarily by cytochrome P450 (CYP) enzymes, particularly CYP3A4.
Side Effects
Contraindications
- •Hypersensitivity to etoricoxib or any component of the formulation
- •Active peptic ulceration or gastrointestinal (GI) bleeding
- •Severe hepatic dysfunction
- •Severe renal dysfunction (CrCl <30 mL/min)
- •Congestive heart failure (NYHA Class II-IV)
- •Established ischemic heart disease, peripheral arterial disease, and/or cerebrovascular disease
- •Uncontrolled hypertension
- •Pregnancy and lactation
Drug Interactions
Lithium
May increase plasma lithium levels, leading to toxicity.
Warfarin
May increase INR and bleeding risk. Close monitoring of INR is recommended.
Diuretics
May reduce the natriuretic effect of loop and thiazide diuretics.
Rifampicin
May decrease etoricoxib plasma concentrations due to enzyme induction.
Methotrexate
May increase methotrexate plasma concentrations, leading to toxicity.
Oral Contraceptives
May increase estrogen and progestin exposure, potentially increasing side effects.
ACE inhibitors/Angiotensin II antagonists
May reduce the antihypertensive effect and increase the risk of renal impairment.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of an overdose, symptomatic and supportive measures should be taken. There is no specific antidote for etoricoxib.
Pregnancy & Lactation
Etoricoxib is contraindicated during pregnancy, especially in the third trimester due to the risk of premature closure of the ductus arteriosus and potential renal dysfunction in the fetus. It is also contraindicated during lactation as it may be excreted in breast milk.
