Description
Overview of the medicine
Flxzen 125 mg Suspension is an antibiotic from the penicillin group, effective against a wide range of bacterial infections. It is particularly useful for infections caused by penicillinase-producing staphylococci.
Uses & Indications
Dosage
Adults
250-500 mg orally every 6 hours, usually taken 30-60 minutes before meals. Dosage may be increased in severe infections.
Elderly
Similar to adult dosage, but caution with renal impairment. Dose reduction may be necessary if creatinine clearance is significantly reduced.
Renal_impairment
For creatinine clearance <10 mL/min, maximum dose 250 mg every 8-12 hours.
How to Take
Oral administration. Shake the bottle well before each use. Take on an empty stomach (at least 30-60 minutes before food or 2 hours after food) for optimal absorption. Use the provided measuring spoon or cup.
Mechanism of Action
Flucloxacillin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to cell lysis and bacterial death. It is resistant to degradation by beta-lactamase (penicillinase) enzymes produced by some bacteria.
Pharmacokinetics
Onset
Within 30-60 minutes
Excretion
Mainly excreted unchanged by the kidneys via tubular secretion and glomerular filtration. A small amount is excreted in bile.
Half life
Approximately 1 hour (in adults with normal renal function)
Absorption
Well absorbed from the gastrointestinal tract, especially when taken on an empty stomach. Peak plasma concentrations are reached within 1-2 hours.
Metabolism
Partially metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to penicillin antibiotics or any component of the suspension.
- •History of flucloxacillin-associated jaundice/hepatic dysfunction.
Drug Interactions
Warfarin
May enhance anticoagulant effect; monitor INR closely.
Probenecid
Increases flucloxacillin plasma concentrations and prolongs its half-life by reducing renal tubular secretion.
Methotrexate
May increase methotrexate toxicity due to reduced renal excretion.
Oral Contraceptives
May reduce efficacy of oral contraceptives (though evidence is limited, advise additional contraception).
Storage
Store dry powder below 30°C. Once reconstituted, store in a refrigerator (2°C-8°C) and discard any unused portion after 7-14 days (check specific product leaflet).
Overdose
Symptoms of overdose may include nausea, vomiting, diarrhea, and electrolyte imbalance. In rare cases, neurological toxicity (e.g., convulsions) may occur, especially in patients with impaired renal function. Treatment is largely supportive; hemodialysis may be considered in severe cases with renal failure.
Pregnancy & Lactation
Pregnancy category B. Generally considered safe during pregnancy, but use only if clearly needed. Excreted in breast milk in small amounts; generally considered compatible with breastfeeding, but monitor infant for potential adverse effects (e.g., diarrhea, candidiasis).
