Overview of the medicine
Gaspro 40 mg capsule is a proton pump inhibitor used to reduce stomach acid production, treating conditions like heartburn, acid reflux, and ulcers caused by excess acid.
Uses & Indications
Dosage
Adults
Usually 20-40 mg once daily, depending on the condition. For severe GERD, 40 mg daily for 4-8 weeks. For H. pylori eradication, 40 mg once or twice daily with antibiotics.
Elderly
No specific dose adjustment is generally required, but caution is advised due to potential for decreased hepatic function.
Renal_impairment
No dose adjustment is necessary for patients with impaired renal function.
How to Take
Take the capsule whole with water at least 30 minutes before a meal, preferably in the morning. Do not chew or crush.
Mechanism of Action
Omeprazole, a proton pump inhibitor, irreversibly blocks the H+/K+ ATPase enzyme system (proton pump) in the gastric parietal cells, thereby reducing gastric acid secretion.

Excretion
Mainly excreted in urine (about 77%) and feces (about 18%) as metabolites.
Half life
0.5-1 hour, but duration of action is longer due to irreversible binding.
Absorption
Rapidly absorbed from the gastrointestinal tract, primarily in the small intestine. Bioavailability is about 30-40%.
Metabolism
Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system, particularly CYP2C19 and CYP3A4.

- Hypersensitivity to omeprazole or any other components of the formulation
- Concomitant use with Nelfinavir

Warfarin
Increased INR/prothrombin time.
Clopidogrel
Reduced antiplatelet effect of clopidogrel.
Nelfinavir, Atazanavir
Reduced plasma concentrations of antiretrovirals.
Diazepam, Phenytoin, Citalopram
Increased plasma levels of these drugs.
