Description
Overview of the medicine
Klavin is an antibiotic containing Amoxicillin and Clavulanic Acid. It is used to treat a wide range of bacterial infections that are resistant to Amoxicillin alone due to beta-lactamase producing bacteria.
Uses & Indications
Dosage
Adults
Usual adult dose is 500 mg amoxicillin/125 mg clavulanic acid three times a day, or 875 mg amoxicillin/125 mg clavulanic acid twice a day, depending on the severity and type of infection.
Elderly
Dosage as for adults, but renal function should be monitored, and dose adjusted if necessary.
Renal_impairment
Dosage should be adjusted based on creatinine clearance (e.g., lower dose or extended dosing interval for severe impairment).
How to Take
Take at the start of a meal to minimize potential gastrointestinal intolerance and enhance absorption. Complete the full course of treatment as prescribed by your doctor.
Mechanism of Action
Amoxicillin works by inhibiting bacterial cell wall synthesis, leading to cell lysis. Clavulanic Acid inhibits beta-lactamase enzymes produced by bacteria, thereby preventing the breakdown of amoxicillin and extending its antibacterial spectrum.

Excretion
Primarily via the kidneys; amoxicillin is excreted largely unchanged, while clavulanic acid is excreted as metabolites and unchanged drug.

Absorption
Both amoxicillin and clavulanic acid are well absorbed from the gastrointestinal tract following oral administration. Peak plasma concentrations are achieved approximately 1-2 hours after dosing.
Metabolism
Amoxicillin is minimally metabolized; clavulanic acid is extensively metabolized to inactive compounds.
Side Effects
Contraindications
- History of hypersensitivity to penicillins or any other beta-lactam antibiotics (e.g., cephalosporins)
- History of cholestatic jaundice or hepatic dysfunction associated with amoxicillin/clavulanic acid therapy
Drug Interactions
Warfarin
Increased INR/prothrombin time, requiring careful monitoring.
Allopurinol
Increased incidence of skin rash when co-administered.
Methotrexate
Reduced renal tubular secretion of methotrexate, potentially increasing its toxicity.

Overdose
