Description
Overview of the medicine
Kofen 1 mg Syrup contains Ketotifen, an antihistamine and mast cell stabilizer, primarily used for the long-term prophylactic treatment of bronchial asthma, allergic conjunctivitis, and other allergic conditions.
Uses & Indications
Dosage
Adults
1 mg (5 ml) twice daily with food. If necessary, the dose may be increased to 2 mg (10 ml) twice daily.
Elderly
No specific dosage adjustment is generally required, but caution is advised due to potential for increased sensitivity.
Renal_impairment

How to Take
Administer orally, usually with food, to minimize gastrointestinal upset. Consistent administration is important for prophylactic effect.
Mechanism of Action
Ketotifen inhibits the release of histamine and other mediators from mast cells and blocks H1-histamine receptors. It also antagonizes the effects of leukotrienes, thereby reducing allergic inflammation.
Pharmacokinetics
Onset
Antihistaminic effect within hours; prophylactic anti-asthmatic effect may take several weeks (2-3 months) to develop fully.
Excretion
Excreted mainly in urine (about 60-70%) as metabolites, with a small amount excreted unchanged.
Half life
Plasma half-life of 12-24 hours for the parent compound, with an active metabolite having a longer half-life.
Absorption
Rapidly absorbed after oral administration. Bioavailability is about 50% due to first-pass metabolism.
Metabolism
Extensively metabolized in the liver, primarily by N-glucuronidation.

Contraindications
- •Hypersensitivity to Ketotifen or any component of the formulation
- •Acute asthma attacks (not for rescue therapy)
