Description
Overview of the medicine
Kuit 1mg Tablet contains Aripiprazole, an atypical antipsychotic medication used to treat various psychiatric conditions including schizophrenia, bipolar I disorder, and as an adjunctive therapy for major depressive disorder.
Uses & Indications
Dosage
Adults
Schizophrenia: Initial 10-15 mg/day, target range 10-30 mg/day. Bipolar I Mania: Initial 15 mg/day. MDD Adjunctive: Initial 2-5 mg/day, target range 5-15 mg/day. Kuit 1mg is typically used for titration or in sensitive patients.
Elderly
Start with lower doses (e.g., 2 mg/day) and titrate gradually based on tolerability and response. Elderly patients with dementia-related psychosis have increased mortality risk.
Renal_impairment
No dosage adjustment is generally required for patients with renal impairment.
How to Take
Kuit 1mg Tablet should be taken orally once daily, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Aripiprazole is thought to work by acting as a partial agonist at dopamine D2 and serotonin 5-HT1A receptors, and as an antagonist at serotonin 5-HT2A receptors. This unique mechanism helps to stabilize dopamine activity in the brain.
Pharmacokinetics
Onset
Clinical improvement typically seen within 1-2 weeks for most indications, but full effects may take longer.
Excretion
Approximately 55% of the dose is excreted in the feces, and 25% is excreted in the urine. Less than 1% is excreted as unchanged drug.
Half life
The elimination half-life of aripiprazole is approximately 75 hours, and its active metabolite, dehydro-aripiprazole, has a half-life of about 94 hours.
Absorption
Well absorbed after oral administration, with peak plasma concentrations typically occurring within 3-5 hours. Absolute oral bioavailability is approximately 87%.
Metabolism
Extensively metabolized in the liver primarily by CYP2D6 and CYP3A4 enzymes. Dehydro-aripiprazole is the only active metabolite.
Side Effects
Contraindications
- •Known hypersensitivity to Aripiprazole or any component of the formulation.
Drug Interactions
CNS depressants (e.g., Alcohol, Benzodiazepines)
Concurrent use may potentiate CNS depressive effects; caution advised.
Strong CYP3A4 inducers (e.g., Carbamazepine, Rifampin)
Double aripiprazole dose.
Strong CYP3A4 inhibitors (e.g., Ketoconazole, Itraconazole)
Reduce aripiprazole dose by half.
Strong CYP2D6 inhibitors (e.g., Quinidine, Fluoxetine, Paroxetine)
Reduce aripiprazole dose by half.
Storage
Store below 30°C in a dry place. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include drowsiness, vomiting, tremor, and tachycardia. Management is primarily symptomatic and supportive, including maintaining a clear airway, ensuring adequate oxygenation and ventilation, and monitoring cardiac function and vital signs. Activated charcoal may be useful.
Pregnancy & Lactation
Pregnancy: Use only if the potential benefit justifies the potential risk to the fetus. Neonates exposed to antipsychotic drugs during the third trimester are at risk for extrapyramidal and/or withdrawal symptoms. Lactation: Aripiprazole is excreted in human milk; caution advised. A decision should be made whether to discontinue nursing or the drug, taking into account the importance of the drug to the mother.
