Overview of the medicine
Lasgrain (Pizotifen) is a serotonin and histamine antagonist primarily used for the prophylactic treatment of various types of vascular headaches, including migraine and cluster headaches. It may also stimulate appetite.
Uses & Indications
Dosage
Adults
Initial dose 0.5 mg at night, gradually increased to 0.5 mg three times daily or 1.5 mg once daily at night. Max 3 mg/day.
Elderly
Same as adults, but caution advised due to increased sensitivity to anticholinergic effects.
Renal_impairment
Dose adjustment may be necessary in severe renal impairment. Use with caution.
How to Take
Oral administration. Can be taken with or without food. To minimize sedation, initial doses should be taken at night.
Mechanism of Action
Pizotifen acts as a serotonin (5-HT1 and 5-HT2) antagonist and a weak H1-receptor antagonist. It is believed to stabilize cranial blood vessels and reduce the inflammatory response associated with migraine. It also has appetite stimulant properties.

Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms include drowsiness, coma, ataxia, tachycardia, hypotension, nausea, vomiting, confusion, and hallucinations. Treatment is symptomatic and supportive, including gastric lavage and activated charcoal.
Pregnancy & Lactation
Pregnancy category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Avoid during lactation as Pizotifen is excreted in breast milk.
