Lipitin 10 mg Tablet is a medication used to lower cholesterol and triglycerides in the blood. It belongs to a group of medicines called statins, which reduce the production of cholesterol by the liver. It helps to prevent heart disease and stroke.
Uses & Indications
Dosage
Adults
Initial dose 10 mg once daily. Dose may be adjusted at 4-week intervals to a maximum of 80 mg once daily based on therapeutic response and tolerability.
Elderly
No specific dosage adjustment required; use lowest effective dose.
Renal_impairment
No dosage adjustment necessary for patients with renal impairment.
How to Take
Take orally once daily, with or without food, at any time of day.
Mechanism of Action
Atorvastatin selectively inhibits HMG-CoA reductase, an enzyme that catalyzes the early, rate-limiting step in cholesterol biosynthesis. This inhibition leads to a decrease in hepatic cholesterol synthesis and, consequently, to an upregulation of hepatic LDL receptors, which increases the uptake and catabolism of LDL-C from the circulation.
Pharmacokinetics
Onset
Significant lipid-lowering effects observed within 2 weeks, maximal effect typically within 4 weeks.
Excretion
Primarily excreted in the bile following hepatic and/or extrahepatic metabolism; less than 2% is recovered in urine.
Half life
Approximately 14 hours for the parent drug, but the half-life of HMG-CoA reductase inhibitory activity is 20-30 hours due to active metabolites.
Absorption
Rapidly absorbed after oral administration; absolute bioavailability is approximately 14%.
Metabolism
Extensively metabolized in the liver by cytochrome P450 3A4 to active ortho- and parahydroxylated metabolites.
Side Effects
Contraindications
- Active liver disease or unexplained persistent elevations of serum transaminases.
- Pregnancy and lactation.
- Hypersensitivity to atorvastatin or any component of the formulation.
Drug Interactions
Colchicine
Cases of myopathy, including rhabdomyolysis, have been reported with coadministration of atorvastatin and colchicine.
Cyclosporine, telaprevir, glecaprevir/pibrentasvir
Significantly increased atorvastatin exposure; avoid coadministration.
Gemfibrozil, other fibrates, niacin (lipid-modifying doses)
Increased risk of myopathy/rhabdomyolysis.
Strong CYP3A4 inhibitors (e.g., clarithromycin, itraconazole, protease inhibitors)
Increased atorvastatin plasma concentrations; limit atorvastatin dose.
Storage
Store at room temperature (below 30°C), protected from light and moisture. Keep out of reach of children.
Overdose
There is no specific treatment for atorvastatin overdose. In the event of an overdose, the patient should be treated symptomatically, and supportive measures instituted as required. Due to extensive drug binding to plasma proteins, hemodialysis is not expected to significantly enhance atorvastatin clearance.
Pregnancy & Lactation
Contraindicated during pregnancy and lactation. Women of childbearing potential should use adequate contraception. If pregnancy occurs, discontinue Lipitin immediately.
