Lipitin 40 mg Tablet contains Atorvastatin, which is used to lower high cholesterol and triglyceride levels in the blood. It helps prevent heart disease, strokes, and other cardiovascular problems.
Uses & Indications
Dosage
Adults
Initial dose is 10 or 20 mg once daily. Dosage can be adjusted based on lipid levels and therapeutic goals, ranging from 10 mg to a maximum of 80 mg once daily.
Elderly
No specific dosage adjustment is required for elderly patients.
Renal_impairment
Dosage adjustment is not required in patients with renal impairment.
How to Take
Take Lipitin 40 mg Tablet orally once daily, with or without food. It can be taken at any time of day, but it is recommended to take it at the same time each day.
Mechanism of Action
Atorvastatin inhibits HMG-CoA reductase, an enzyme crucial for cholesterol synthesis in the liver. By reducing cholesterol production, it decreases LDL (bad cholesterol) and VLDL levels, while increasing HDL (good cholesterol).
Pharmacokinetics
Onset
Lipid-lowering effects typically begin within 2 weeks, with maximal response usually observed by 4 weeks.
Excretion
Primarily eliminated in the bile following hepatic and/or extrahepatic metabolism. Less than 2% of a dose is recovered in the urine.
Half life
The apparent plasma elimination half-life of atorvastatin is approximately 14 hours, but the half-life of HMG-CoA reductase inhibitory activity is 20 to 30 hours due to active metabolites.
Absorption
Rapidly absorbed after oral administration; peak plasma concentrations reached within 1 to 2 hours. Absolute bioavailability is approximately 12% due to first-pass metabolism.
Metabolism
Extensively metabolized in the liver by cytochrome P450 3A4 (CYP3A4) to ortho- and parahydroxylated derivatives and various beta-oxidation products.
Side Effects
Contraindications
- Active liver disease or unexplained persistent elevations of serum transaminases
- Pregnancy and lactation
- Hypersensitivity to atorvastatin or any component of the formulation
Drug Interactions
Niacin
Increased risk of myopathy/rhabdomyolysis.
Cyclosporine
Coadministration significantly increases atorvastatin plasma concentrations, increasing the risk of myopathy.
Oral contraceptives
May increase plasma concentrations of norethindrone and ethinyl estradiol.
Gemfibrozil and other fibrates
Increased risk of myopathy when coadministered with atorvastatin.
Macrolide antibiotics (e.g., Clarithromycin)
Increased exposure to atorvastatin and increased risk of myopathy/rhabdomyolysis.

There is no specific treatment for atorvastatin overdose. In the event of an overdose, patients should be treated symptomatically and supportive measures instituted as required.
