Description
Overview of the medicine
Natazol (Natamycin) is a broad-spectrum antifungal agent primarily used for topical treatment of fungal infections of the eye, including fungal keratitis, conjunctivitis, and blepharitis caused by susceptible fungi.
Uses & Indications
Dosage
Adults
For fungal keratitis, 1 drop into the affected eye(s) every 1-2 hours for 3-4 days, then 1 drop 6-8 times daily for up to 14-21 days or until resolution. Dosage may be tapered gradually.
Elderly
No specific dose adjustment is required for elderly patients.
Renal_impairment
No dose adjustment is necessary due to minimal systemic absorption.
How to Take
Shake well before use. Instill drops into the conjunctival sac. Avoid touching the dropper tip to the eye or any other surface to prevent contamination. If using other eye drops, wait at least 5-10 minutes between applications.
Mechanism of Action
Natamycin acts by binding to ergosterol, a vital component of fungal cell membranes, leading to increased permeability of the membrane. This disruption causes leakage of essential intracellular components and ultimately fungal cell death.
Pharmacokinetics
Onset
Clinical improvement may be seen within days, but full eradication takes weeks.
Excretion
Not relevant for topical ophthalmic use due to negligible systemic absorption.
Half life
Not relevant for topical ophthalmic use due to negligible systemic absorption.
Absorption
Minimal systemic absorption occurs after topical ophthalmic administration due to its poor solubility and large molecular size. Primarily acts locally on the eye surface.
Metabolism
No significant systemic metabolism due to minimal absorption.
Side Effects
Contraindications
- Hypersensitivity to Natamycin or any component of the formulation.
Drug Interactions
Other topical ophthalmic medications
Allow at least 5-10 minutes between applications of different eye drops to prevent wash-out and ensure adequate absorption.
Storage
Store at controlled room temperature (20-25°C). Protect from light. Do not freeze. Keep out of reach of children.
Overdose
Topical ophthalmic overdose is unlikely to cause systemic toxicity due to minimal absorption. Excessive local application may lead to ocular irritation, which can be managed by rinsing the eye with sterile water.
Pregnancy & Lactation
Pregnancy Category C. Use only if clearly needed. Minimal systemic absorption suggests low risk, but caution is advised. Consult a doctor before use during pregnancy or lactation.
