Nauset 4 mg Oral Solution contains Ondansetron, an antiemetic medication used to prevent nausea and vomiting caused by chemotherapy, radiation therapy, and surgery. It works by blocking a natural substance (serotonin) in the body that causes nausea and vomiting.
Uses & Indications
Dosage
Adults
Chemotherapy-induced nausea and vomiting: For highly emetogenic chemotherapy, 8 mg 30 minutes before chemotherapy, then 8 mg 8 hours after the first dose, followed by 8 mg twice daily for 1-2 days. For moderately emetogenic chemotherapy, 8 mg 30 minutes before chemotherapy, then 8 mg 8-12 hours after the first dose, or 8 mg twice daily for 1-2 days. Post-operative nausea and vomiting: 4 mg as a single dose.
Elderly
No dosage adjustment is generally required for patients over 65 years, except for severe hepatic impairment.
Renal_impairment
No dosage adjustment is required for patients with renal impairment.
Hepatic_impairment
In patients with severe hepatic impairment, the total daily dose should not exceed 8 mg.
How to Take
Take Nauset oral solution by mouth, usually with or without food, as directed by your doctor. Measure the dose carefully using the provided measuring cup or oral syringe. Do not exceed the prescribed dose.
Mechanism of Action
Ondansetron is a highly selective antagonist of 5-HT3 receptors. Chemotherapeutic agents and radiation therapy cause release of serotonin from enterochromaffin cells of the small intestine. This serotonin activates 5-HT3 receptors located on vagal afferents, initiating the vomiting reflex. Ondansetron blocks this activation, thereby preventing nausea and vomiting.

Within 30 minutes to 2 hours.
Excretion
Approximately 5% of the dose is excreted unchanged in urine; the remainder is excreted as metabolites via urine and feces.
Half life
Approximately 3-5 hours in adults, longer in elderly and those with hepatic impairment.
Absorption
Rapidly and completely absorbed from the gastrointestinal tract following oral administration, but undergoes some first-pass metabolism.
Metabolism
Extensively metabolized in the liver by multiple cytochrome P450 enzymes (CYP1A2, CYP2D6, CYP3A4) to inactive metabolites.

Contraindications
- Hypersensitivity to Ondansetron or any component of the formulation.
- Concomitant use with apomorphine due to the risk of profound hypotension and loss of consciousness.
Drug Interactions
Tramadol
Ondansetron may reduce the analgesic effect of tramadol.
Apomorphine
Concomitant use is contraindicated due to reports of profound hypotension and loss of consciousness.
Drugs that prolong QT interval
Caution should be exercised when co-administering ondansetron with other drugs that prolong the QT interval (e.g., antiarrhythmics, antipsychotics).
