Nauset 8 mg Injection contains Ondansetron, an anti-emetic drug used to prevent nausea and vomiting caused by chemotherapy, radiation therapy, and surgery.
Uses & Indications
Dosage
Adults
Chemotherapy-induced nausea and vomiting (CINV): Highly emetogenic: 24 mg orally once, 30 minutes before chemotherapy, or 8 mg IV/IM immediately before chemotherapy, followed by 8 mg orally twice daily for 1-2 days. Moderately emetogenic: 8 mg orally twice daily or 8 mg IV/IM once immediately before chemotherapy. Post-operative nausea and vomiting (PONV): 4 mg IV/IM as a single dose immediately before induction of anesthesia, or post-operatively if nausea/vomiting occurs.
Elderly
No dosage adjustment is generally required for patients over 65 years. However, for patients >75 years, the initial IV dose should not exceed 16 mg.
Renal_impairment
No dosage adjustment is required for patients with renal impairment.
How to Take
Nauset 8 mg Injection is administered intravenously (IV) or intramuscularly (IM) by a healthcare professional. It can be given as a slow IV injection over at least 30 seconds, preferably over 2-5 minutes, or as an IM injection. It may be diluted and infused over 15 minutes for some indications.
Mechanism of Action
Ondansetron is a selective serotonin 5-HT3 receptor antagonist. Serotonin 5-HT3 receptors are present both peripherally on vagal nerve terminals and centrally in the chemoreceptor trigger zone (CTZ). By blocking these receptors, ondansetron prevents the initiation of the vomiting reflex.

Absorption
Rapidly and extensively absorbed after oral administration, but bioavailability is approximately 60% due to first-pass metabolism. IV administration bypasses this.
Metabolism
Extensively metabolized in the liver by cytochrome P450 enzymes (CYP1A2, CYP2D6, CYP3A4) via hydroxylation, followed by glucuronide or sulfate conjugation.
Side Effects
Contraindications
- Hypersensitivity to ondansetron or any component of the formulation.
- Concomitant use with apomorphine.
Drug Interactions
Tramadol
May reduce the analgesic effect of tramadol and increase the risk of serotonin syndrome.
Apomorphine
Concomitant use is contraindicated due to reports of profound hypotension and loss of consciousness.
Drugs that prolong QT interval
Use with caution due to potential additive QT prolongation effect (e.g., antiarrhythmics, antipsychotics).
Phenytoin, Carbamazepine, Rifampicin
May decrease ondansetron plasma concentrations.

Symptoms of overdose include transient visual disturbances, severe constipation, hypotension, and vasovagal episodes with transient AV block. ECG monitoring is recommended. Treatment is supportive and symptomatic.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of impaired fertility or harm to the fetus. However, human data is limited. Use during pregnancy only if clearly needed. Ondansetron is excreted in the milk of lactating rats; it is not known whether it is excreted in human milk. Exercise caution when administering to a nursing mother.
