Description
Overview of the medicine
Navalexin 125 mg Suspension is an antibiotic from the cephalosporin group, used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
Usually 250-1000 mg every 6-12 hours depending on infection severity; for 125mg suspension, dosage will be determined by physician for appropriate volume.
Elderly
Dosage adjustment may be necessary in patients with significant renal impairment.
Children
General dose 25-50 mg/kg/day in divided doses (every 6 or 8 hours). For severe infections, up to 100 mg/kg/day, not to exceed 4 g/day.
Renal_impairment
Dosage reduction required based on creatinine clearance (e.g., for CrCl <10 mL/min, 250 mg every 12 hours).
How to Take
Oral, with or without food. Administer with a measuring spoon or cup. Shake the bottle well before each use.
Mechanism of Action
Navalexin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Clinical effects usually seen within a few hours of the first dose.
Excretion
Excreted primarily by the kidneys via glomerular filtration and tubular secretion, with 70-80% of the dose excreted unchanged in urine within 8 hours.
Half life
Approximately 0.5-1.2 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract; peak plasma concentrations reached within 1 hour.
Metabolism
Not significantly metabolized in the liver.
Side Effects
Contraindications
- Known hypersensitivity to cephalosporin antibiotics.
- History of severe hypersensitivity reactions (e.g., anaphylaxis) to penicillin or other beta-lactam antibiotics (due to potential cross-reactivity).
Drug Interactions
Metformin
Cephalexin may increase metformin concentrations; monitor for metformin-related adverse effects.
Probenecid
May increase cephalexin plasma concentrations and prolong its half-life by inhibiting renal excretion.
Other nephrotoxic drugs
Concurrent use should be monitored for potential increased renal toxicity.
Oral anticoagulants (e.g., Warfarin)
May enhance the anticoagulant effect, requiring closer INR monitoring.
Storage
Store the dry powder below 30°C in a dry place, protected from light and moisture. Keep the reconstituted suspension in a refrigerator (2-8°C). Do not freeze. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric distress, diarrhea, and hematuria. Management is supportive, including gastric lavage or activated charcoal. Hemodialysis may be useful in severe cases or in patients with impaired renal function.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe during pregnancy, but should be used only if clearly needed. Excreted in small amounts in breast milk; generally considered compatible with breastfeeding, but monitor infants for diarrhea, candidiasis, or rash.

Cephalexin has been extensively studied since its introduction and has a well-established efficacy and safety profile. Ongoing research focuses on its use in emerging resistance patterns and specific patient populations.
Lab Monitoring
- Renal function tests (blood urea nitrogen and creatinine) should be performed before and during therapy in patients with impaired renal function or receiving prolonged treatment.
- Complete blood count (CBC) should be monitored occasionally during prolonged therapy.
Doctor Notes
- Always inquire about previous allergic reactions, especially to penicillin, before prescribing.
- Emphasize the importance of completing the full course of antibiotics to prevent bacterial resistance and recurrence.
- Adjust dosage in patients with renal impairment based on creatinine clearance.
- Advise parents on proper measurement and storage of the suspension.
Patient Guidelines
- Complete the entire course of medication as prescribed, even if symptoms improve or disappear.
- Shake the suspension well before each use to ensure proper mixing.
- Use the provided measuring device (spoon or cup) for accurate dosing, do not use household spoons.
