Overview of the medicine
Neurolin CR contains Pregabalin, an extended-release medication used to treat neuropathic pain, fibromyalgia, and as an adjunctive therapy for partial onset seizures in adults. Its controlled-release formulation ensures prolonged therapeutic effect and less frequent dosing.
Uses & Indications
Dosage
Adults
Neuropathic pain: Initially 75 mg once daily, adjusted based on response up to 300 mg once daily. Fibromyalgia: Initially 75 mg once daily, adjusted up to 300 mg once daily.
Elderly
Dosage should be adjusted cautiously, especially if renal function is impaired.
Renal_impairment
Requires significant dose reduction based on creatinine clearance.
How to Take
Take orally once daily, with or without food. Swallow whole; do not chew, crush, or split.
Mechanism of Action
Pregabalin binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, reducing the release of several excitatory neurotransmitters, thereby exerting analgesic and anxiolytic effects.

Onset
Pain relief may be noticed within a week; full effect may take longer.
Excretion
Primarily excreted unchanged in the urine.

Approximately 6.3 hours.
Absorption
Well absorbed from the gastrointestinal tract; Cmax reached within 3-4 hours for CR formulation.

Minimally metabolized; less than 0.1% excreted as metabolites.

Contraindications

Drug Interactions
Thiazolidinedione antidiabetic agents
May increase risk of weight gain and peripheral edema.
CNS depressants (opioids, benzodiazepines, alcohol)
Increased risk of sedation and respiratory depression.
Storage
Store below 30°C in a dry place, away from light. Keep out of reach of children.
Overdose
Symptoms include somnolence, confusion, agitation, and restlessness. Management is supportive, including gastric lavage if recent ingestion, and monitoring vital signs.
Pregnancy & Lactation
Pregnancy Category C. Use only if potential benefit justifies potential risk to the fetus. Not recommended during breastfeeding as it is excreted in breast milk.
