Description
Overview of the medicine
Omeprazole is a proton pump inhibitor (PPI) that works by decreasing the amount of acid your stomach makes. It is used to treat heartburn, stomach ulcers, and gastroesophageal reflux disease (GERD).
Uses & Indications
Dosage
Adults
Usually 20 mg once daily for 4-8 weeks, depending on the condition. For severe conditions, it may be increased to 40 mg once daily. For H. pylori eradication, 20 mg twice daily with antibiotics for 7-14 days.
Elderly
No specific dose adjustment is generally required, but caution is advised in patients with severe hepatic impairment.
Renal_impairment
No dose adjustment is necessary for patients with impaired renal function.
How to Take
Take the capsule whole with water, preferably in the morning before a meal. Do not chew or crush the capsule.
Mechanism of Action
Omeprazole irreversibly inhibits the H+/K+-ATPase (proton pump) in the secretory canaliculi of the gastric parietal cells. This reduces gastric acid secretion.
Pharmacokinetics
Onset
Within 1-2 hours.
Excretion
Approximately 80% is excreted in urine as metabolites, and the remainder in feces.
Half life
Plasma elimination half-life is 0.5-1 hour, but the duration of acid suppression is much longer due to irreversible binding.
Absorption
Rapidly absorbed from the gastrointestinal tract, but absorption is variable. Bioavailability increases with repeated administration. Food may delay absorption.
Metabolism
Extensively metabolized in the liver by the cytochrome P450 enzyme system, primarily by CYP2C19 and CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to omeprazole, substituted benzimidazoles, or any component of the formulation.
- Concomitant use with Nelfinavir.
Drug Interactions
Diazepam
Decreased clearance of diazepam.
Warfarin
Increased INR/prothrombin time.
Clopidogrel
Omeprazole may reduce the antiplatelet effect of Clopidogrel.
Methotrexate
Increased plasma levels of methotrexate.
Nelfinavir/Atazanavir
Reduced plasma concentrations of these antiviral drugs.

Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
