Description
Overview of the medicine
Omeprazole is a proton pump inhibitor (PPI) that decreases the amount of acid produced in the stomach. It is used to treat conditions such as gastroesophageal reflux disease (GERD), peptic ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
GERD: 20-40 mg once daily for 4-8 weeks. Duodenal ulcer: 20 mg once daily for 2-4 weeks. Gastric ulcer: 20 mg once daily for 4-8 weeks. H. pylori eradication: 20 mg twice daily with antibiotics. Zollinger-Ellison syndrome: Initial 60 mg once daily, adjusted as needed.
Elderly
No specific dosage adjustment usually required, but caution with hepatic impairment.
Renal_impairment
No dosage adjustment needed for renal impairment.
How to Take
For oral use. Take the capsule whole with water, preferably in the morning before a meal. Do not chew or crush the capsule.
Mechanism of Action
Omeprazole irreversibly binds to and inhibits the H+/K+-ATPase (proton pump) in the secretory canaliculi of parietal cells in the stomach. This blockade of the proton pump prevents the secretion of hydrogen ions into the gastric lumen, thereby reducing gastric acid secretion.

Onset
Within 1-2 hours, maximal effect after 2-4 days of continuous treatment.
Excretion
Mainly via urine (approx. 77%), remainder via feces.

0.5-1 hour (terminal elimination half-life is longer due to irreversible binding).
