Description
Overview of the medicine
Pregabid 75 mg Capsule contains Pregabalin, an anticonvulsant and neuropathic pain agent. It is used to treat neuropathic pain (e.g., diabetic neuropathy, post-herpetic neuralgia), fibromyalgia, and as an adjunctive therapy for partial-onset seizures in adults.
Uses & Indications
Dosage
Adults
Initial dose is usually 75 mg twice daily or 50 mg three times daily (150 mg/day). May be increased to 150 mg twice daily or 100 mg three times daily (300 mg/day) within one week based on efficacy and tolerability. Maximum dose is usually 300 mg twice daily or 200 mg three times daily (600 mg/day).
Elderly
Dose adjustment may be required due to age-related decrease in renal function. Start with a lower dose and titrate cautiously.
Renal_impairment
Dose must be adjusted based on creatinine clearance. For CrCl 30-60 mL/min, maximum daily dose is 300 mg (divided). For CrCl 15-30 mL/min, maximum daily dose is 150 mg (divided). For CrCl <15 mL/min, maximum daily dose is 75 mg (divided).
How to Take
Take orally, with or without food. Swallow the capsule whole with water. Do not crush, chew, or open the capsule.
Mechanism of Action
Pregabalin binds with high affinity to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system. This binding modulates the release of several excitatory neurotransmitters, thereby reducing neuronal hyperexcitability and pain signaling.
Pharmacokinetics
Onset
Pain relief may be seen within days to weeks; antiepileptic effects also observed over a similar timeframe. Anxiolytic effects may appear earlier.
Excretion
Excreted almost entirely unchanged in the urine (approximately 98% of the administered dose). Renal clearance is proportional to creatinine clearance.
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed following oral administration, with peak plasma concentrations occurring within 1.5 hours. Oral bioavailability is ≥90% and is not affected by food.
Metabolism
Negligible systemic metabolism in humans. Less than 0.1% of the dose is excreted as metabolites.

May increase the risk of angioedema when co-administered with pregabalin.
CNS Depressants (e.g., opioids, benzodiazepines, alcohol)
