Description
Overview of the medicine
Pregalex 150 mg Capsule contains Pregabalin, an anticonvulsant and neuropathic pain agent. It is primarily used to treat neuropathic pain, epilepsy, and generalized anxiety disorder.
Uses & Indications
Dosage
Adults
For neuropathic pain: Initially 75 mg twice daily or 50 mg three times daily, adjusted up to 150-300 mg twice daily based on response. For GAD: Initially 150 mg/day in 2-3 divided doses, adjusted up to 300-600 mg/day. For epilepsy: Initially 75 mg twice daily, adjusted up to 600 mg/day.
Elderly
Dosage adjustment may be necessary due to potential age-related decrease in renal function. Start with a lower dose and titrate cautiously.
Renal_impairment
Dosage must be significantly reduced in patients with impaired renal function based on creatinine clearance (e.g., for CrCl <15 mL/min, 25-50 mg once daily or every other day).
How to Take
Swallow whole with water, with or without food. Can be taken as a single dose or in divided doses daily as prescribed by the physician.
Mechanism of Action
Pregabalin binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, which leads to a reduction in the release of several neurotransmitters like glutamate, noradrenaline, and substance P. This action is believed to be responsible for its anticonvulsant, anxiolytic, and analgesic effects.

Onset
Onset of pain relief can be observed within 1 week for neuropathic pain. Full therapeutic effect may take longer.
Excretion
Primarily excreted unchanged in the urine; renal excretion is the major route of elimination.
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations occur within 1.5 hours. Bioavailability is >90% and is not affected by food.
Metabolism
Undergoes negligible metabolism in humans (less than 2% of a dose is metabolized).
Side Effects
Contraindications
- •Known hypersensitivity to pregabalin or any component of the formulation.
- •Patients with rare hereditary problems of galactose intolerance, the Lapp lactase deficiency or glucose-galactose malabsorption should not take this medicine.
Drug Interactions
CNS depressants (e.g., opioids, benzodiazepines, ethanol)
May potentiate CNS depressant effects, leading to increased sedation and respiratory depression. Use with caution.
Thiazolidinedione antidiabetics (e.g., rosiglitazone, pioglitazone)
May increase the risk of peripheral edema and weight gain when co-administered. Closely monitor for fluid retention.
Storage
Store below 30°C, in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusional state, agitation, and rarely coma. Treatment is supportive and symptomatic; hemodialysis can be used if necessary.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Not recommended for use during breastfeeding as pregabalin is excreted in breast milk.
