Description
Overview of the medicine
Rupatadine is a non-sedating, long-acting, selective H1-receptor antagonist used for the symptomatic treatment of allergic rhinitis and urticaria.
Uses & Indications
Dosage
Adults
Adults and adolescents (12 years and older): 10 mg once daily.
Elderly
No dose adjustment is required in elderly patients, but caution is advised due to potential age-related decrease in renal/hepatic function.
Renal_impairment
Use with caution in patients with renal impairment. No specific dose adjustment guidelines for severe renal impairment.
How to Take
Oral administration, with or without food.
Mechanism of Action
Rupatadine is a second-generation H1-antihistamine with selective peripheral H1-receptor antagonist activity. It also inhibits platelet-activating factor (PAF) and mast cell degranulation.

Onset of action within 1-2 hours.
Excretion
Eliminated primarily via feces (60.9%) and urine (34.6%).
Half life
Terminal half-life is approximately 5.9 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma levels reached in 0.75-1.5 hours. Low oral bioavailability (approx. 10%) due to extensive first-pass metabolism.
Metabolism
Extensively metabolized in the liver, mainly by cytochrome P450 (CYP3A4) into active and inactive metabolites.
