Description
Overview of the medicine
Ceftazidime is a third-generation cephalosporin antibiotic used to treat a variety of bacterial infections. It is effective against a broad spectrum of gram-negative bacteria, including Pseudomonas aeruginosa, and some gram-positive organisms. It is administered parenterally, typically intravenously or intramuscularly.
Uses & Indications
Dosage
Adults
Typically 1-2 g intravenously or intramuscularly every 8-12 hours, depending on the severity of the infection and susceptibility of the pathogen. Maximum dose up to 6 g/day.
Elderly
No specific dose adjustment for elderly patients with normal renal function. Doses should be adjusted based on renal function.
Renal_impairment
Dosage must be adjusted based on creatinine clearance. For severe impairment, dose may be significantly reduced and/or interval extended.
How to Take
Administer by deep intramuscular injection or by slow intravenous injection (3-5 minutes) or by intravenous infusion (20-30 minutes). Reconstitute the powder with sterile water for injection, according to manufacturer's instructions.
Mechanism of Action
Ceftazidime is a bactericidal antibiotic that acts by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) located in the bacterial cytoplasmic membrane, thereby interfering with the transpeptidation step of peptidoglycan synthesis. This leads to disruption of the cell wall, ultimately causing bacterial cell lysis and death.

Excretion
Primarily excreted unchanged by the kidneys (glomerular filtration and tubular secretion).
Half life
Approximately 1.8-2 hours in adults with normal renal function.
Absorption
Rapid and complete absorption after intramuscular administration; peak plasma concentrations reached within approximately 1 hour. Intravenous administration results in immediate peak concentrations.

Not metabolized; excreted unchanged.

Contraindications
- Hypersensitivity to ceftazidime, other cephalosporins, or any component of the formulation.
- History of severe hypersensitivity reaction to any other type of beta-lactam antibacterial agent (e.g., penicillins, carbapenems).

Furosemide
Concomitant use with potent diuretics like furosemide may enhance nephrotoxicity.
Aminoglycosides
May increase nephrotoxicity, especially in patients with pre-existing renal impairment.
