How to Take
Administer orally. Use the provided dropper for accurate dosing. Shake the bottle well before use. Can be given with or without food.
Mechanism of Action
Paracetamol's exact mechanism is not fully understood but is believed to involve inhibition of prostaglandin synthesis in the central nervous system (CNS). It has weak anti-inflammatory effects.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Mainly excreted in the urine as glucuronide and sulfate conjugates, with a small amount of unmetabolized paracetamol.
Half life
1.5-3 hours in adults; slightly longer in neonates and shorter in children.
Absorption
Rapidly and almost completely absorbed from the gastrointestinal tract.
Metabolism
Primarily metabolized in the liver via glucuronidation and sulfation, to a lesser extent by cytochrome P450 to a highly reactive intermediate (NAPQI), which is detoxified by glutathione.
Side Effects
Contraindications
- Hypersensitivity to paracetamol or any component of the formulation.
- Severe hepatic impairment or active liver disease.
Drug Interactions
Alcohol
Increases risk of hepatotoxicity.
Warfarin
May increase anticoagulant effect (risk of bleeding) with prolonged regular use.
Cholestyramine
Reduces paracetamol absorption if given within one hour.
Phenytoin, Phenobarbital, Carbamazepine
May increase paracetamol metabolism, potentially reducing efficacy and increasing hepatotoxicity.
Storage
Store below 30°C in a dry place, protected from light. Do not freeze. Keep away from direct sunlight.
Overdose
Symptoms include nausea, vomiting, abdominal pain, pallor, and anorexia. Severe overdose can lead to hepatotoxicity (liver damage), which may be delayed for 24-48 hours. Management involves gastric lavage, activated charcoal, and administration of N-acetylcysteine (NAC) as an antidote.
Pregnancy & Lactation
Considered safe for use during pregnancy and lactation when used at recommended doses, as it is widely used and well-tolerated. Always consult a healthcare professional.
