Description
Overview of the medicine
Urodin (Solifenacin Succinate) is used to treat symptoms of overactive bladder (OAB) such as urinary urgency, frequency, and urge incontinence.
Uses & Indications
Dosage
Adults
The recommended starting dose is 5 mg once daily. If needed, the dose may be increased to 10 mg once daily.
Elderly
No specific dose adjustment is required for elderly patients.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), the maximum dose should be 5 mg once daily.
Hepatic_impairment
For patients with moderate hepatic impairment (Child-Pugh B), the maximum dose should be 5 mg once daily. Not recommended for severe hepatic impairment (Child-Pugh C).
How to Take
Take orally with water, with or without food. Swallow the tablet whole; do not crush or chew.
Mechanism of Action
It is a competitive muscarinic receptor antagonist that selectively blocks M3 receptors in the bladder, thereby reducing involuntary contractions of the detrusor muscle.
Pharmacokinetics
Onset
Clinical effects are typically observed within one week of starting treatment, with full efficacy seen within 4-8 weeks.
Excretion
Approximately 69% of the dose is excreted in the urine and 23% in the feces.
Half life
The elimination half-life is approximately 45-68 hours.
Absorption
Well absorbed after oral administration, bioavailability is approximately 90%. Peak plasma concentrations (Tmax) are reached within 3 to 8 hours.
Metabolism
Extensively metabolized in the liver, primarily by the cytochrome P450 3A4 (CYP3A4) enzyme system.
Side Effects
Contraindications
- Urinary retention
- Gastric retention
- Uncontrolled narrow-angle glaucoma
- Severe hepatic impairment (Child-Pugh C)
- Patients undergoing hemodialysis
- Known hypersensitivity to solifenacin succinate or any component of the formulation.
Drug Interactions
0
Potent CYP3A4 inhibitors (e.g., ketoconazole, ritonavir): Reduce Urodin dose to 5 mg.
1
CYP3A4 inducers (e.g., rifampicin, phenytoin): May decrease solifenacin exposure.
2
Other anticholinergic drugs: May enhance anticholinergic side effects.
3
Cholinergic receptor agonists: May reduce the effect of cholinergic agonists.
4
Drugs that prolong QT interval: Use with caution as solifenacin may prolong QT interval.
Storage
Store below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include severe anticholinergic effects such as dry mouth, blurred vision, constipation, urinary retention, tachycardia, and mydriasis. Treatment should be symptomatic and supportive, including gastric lavage, activated charcoal, and if severe CNS effects occur, physostigmine.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown if solifenacin is excreted in human milk, so caution should be exercised when administered to a nursing mother.

Doctor Notes
- Advise patients on lifestyle modifications.
- Monitor for anticholinergic side effects, especially in elderly and those with pre-existing conditions (e.g., glaucoma, cardiac issues).
- Consider dose reduction in patients with moderate renal or hepatic impairment.
