Description
Overview of the medicine
Velofix 100 mg Suspension is an antibiotic medication containing Cefixime, primarily used to treat various bacterial infections in children. It belongs to the cephalosporin class of antibiotics and works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Adults: 400 mg daily as a single dose or in two divided doses (not common for 100 mg suspension, typically used for pediatric patients or by conversion from higher strength forms).
Elderly
Similar to adults; dosage adjustment may be necessary in patients with significant renal impairment.
Renal_impairment
Creatinine clearance 21-60 mL/min: 75% of standard dose. Creatinine clearance <20 mL/min: 50% of standard dose.
How to Take
Velofix 100 mg Suspension is for oral administration. It can be taken with or without food. Shake the bottle well before each use. Use the provided measuring device (e.g., spoon or syringe) to ensure accurate dosing.
Mechanism of Action
Cefixime is a bactericidal antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), thereby interfering with the cross-linking of peptidoglycan chains. This leads to the disruption of the bacterial cell wall and ultimately bacterial cell lysis and death.
Pharmacokinetics
Onset
Antibacterial effects begin rapidly, within a few hours of administration.
Excretion
Primarily excreted unchanged by the kidneys (glomerular filtration and tubular secretion). About 10% is eliminated via bile.
Half life
Approximately 3-4 hours.
Absorption
Approximately 40-50% orally absorbed. Peak serum concentrations are achieved within 2-6 hours.
Metabolism
Not extensively metabolized in the body. About 50% of the absorbed dose is excreted unchanged in the urine.
Side Effects
Contraindications
- Hypersensitivity to Cefixime or any other cephalosporin antibiotics.
- History of serious hypersensitivity reactions to penicillin or other beta-lactam antibiotics.
Drug Interactions
Warfarin
May increase prothrombin time (PT) and INR, increasing the risk of bleeding. Close monitoring is recommended.
Nifedipine
May increase the bioavailability of cefixime.
Carbamazepine
Concomitant use may increase carbamazepine levels. Therapeutic drug monitoring may be necessary.
Oral Contraceptives
May reduce the efficacy of oral contraceptives in some cases, consider additional birth control methods.
Storage
Store the dry powder below 30°C, protected from light and moisture. After reconstitution, store the liquid suspension in a refrigerator (2-8°C) and discard any unused portion after the recommended period.
Overdose
In case of overdose, symptomatic and supportive treatment is recommended. Gastric lavage may be indicated. Cefixime is not removed in significant quantities by dialysis.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown harm to the fetus, but there are no adequate and well-controlled studies in pregnant women. Use during pregnancy only if clearly needed. Cefixime is excreted in human milk; caution should be exercised when administered to a nursing woman.
