Overview of the medicine
Vigorex 100 mg Tablet contains Sildenafil Citrate, which is used to treat erectile dysfunction (impotence) in men. It helps to achieve and maintain an erection sufficient for sexual activity by increasing blood flow to the penis.
Uses & Indications
Dosage
Adults
The recommended dose is 50 mg taken approximately 1 hour before sexual activity. Based on efficacy and tolerability, the dose may be increased to 100 mg or decreased to 25 mg. The maximum recommended dosing frequency is once per day.
Elderly
No dosage adjustment is required for healthy elderly patients (≥65 years of age).
Renal_impairment
Mild to moderate renal impairment (creatinine clearance 30-80 mL/min): No dosage adjustment needed. Severe renal impairment (creatinine clearance <30 mL/min): Consider a 25 mg initial dose.
How to Take
For oral administration. Vigorex 100 mg Tablet can be taken with or without food. However, taking it with a high-fat meal may delay the onset of action.
Mechanism of Action
Sildenafil is a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5). When sexual stimulation causes local release of nitric oxide, PDE5 inhibition by sildenafil increases cGMP levels in the corpus cavernosum, leading to smooth muscle relaxation and inflow of blood into the penis.
Pharmacokinetics
Onset
Typically 30-60 minutes after oral administration.
Excretion
Excreted predominantly as metabolites in the feces (approximately 80% of the administered oral dose) and to a lesser extent in the urine (approximately 13% of the administered oral dose).
Half life
Approximately 3-5 hours for sildenafil and about 4 hours for its active metabolite, N-desmethyl sildenafil.
Absorption
Rapidly absorbed after oral administration. Mean absolute bioavailability is about 41%. Peak plasma concentrations are reached within 30-120 minutes (median 60 minutes) of oral dosing in the fasted state. When taken with a high-fat meal, the rate of absorption is reduced.
Metabolism
Primarily metabolized by hepatic microsomal isoenzymes CYP3A4 (major route) and CYP2C9 (minor route). The major circulating metabolite, N-desmethyl sildenafil, has about 50% of the potency of the parent drug.
Side Effects
Contraindications
- Concomitant use with nitrates or nitric oxide donors (e.g., nitroglycerin, isosorbide dinitrate) in any form, due to potentiation of hypotensive effects.
- Known hypersensitivity to sildenafil or any component of the tablet.
- Patients with severe cardiovascular disease for whom sexual activity is inadvisable.
- Recent myocardial infarction (within the last 90 days) or stroke (within the last 6 months).
- Severe hepatic impairment.
- Known hereditary degenerative retinal disorders, such as retinitis pigmentosa.
Drug Interactions
Nitrates
Potentiation of hypotensive effects; concomitant use is contraindicated.
Riociguat
Concomitant use is contraindicated due to additive hypotensive effects.
Alpha-blockers
Risk of symptomatic hypotension, especially with initial dosing and titration.
CYP3A4 Inhibitors (e.g., Ritonavir, Ketoconazole, Erythromycin)
Increased plasma concentrations of sildenafil, leading to a higher risk of side effects. Dose adjustment may be necessary.

Store below 30°C in a cool, dry place, away from light and moisture. Keep out of reach of children.
Overdose
In studies with healthy volunteers, doses up to 800 mg have been given. Adverse reactions were similar to those seen at lower doses but generally at an increased rate and severity. In cases of overdose, standard supportive measures should be adopted as required. Renal dialysis is not expected to accelerate clearance as sildenafil is highly bound to plasma proteins and not eliminated in the urine.
Pregnancy & Lactation
Vigorex 100 mg Tablet is not indicated for use in women and therefore should not be used during pregnancy or lactation.
