Vigorex 25 mg Tablet is a medication used to treat erectile dysfunction (impotence) in men. It helps to achieve and maintain an erection sufficient for sexual activity by increasing blood flow to the penis.
Uses & Indications
Dosage
Adults
For most patients, the starting dose is 25 mg taken orally, as needed, approximately 1 hour before sexual activity. It may be taken between 30 minutes to 4 hours before sexual activity. The dose may be adjusted based on individual response and tolerability. Do not exceed one dose per 24 hours.
Elderly
No dose adjustment is generally required, but consideration should be given to a lower starting dose (e.g., 25 mg) due to potential age-related decrease in clearance and increased sensitivity to side effects.
Renal_impairment
For severe renal impairment (creatinine clearance <30 mL/min), a starting dose of 25 mg should be considered.
How to Take
Take Vigorex 25 mg Tablet orally with a glass of water, approximately 30-60 minutes before sexual activity. It can be taken with or without food, but a high-fat meal may delay the onset of action.
Mechanism of Action
Sildenafil is a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5). It enhances the effects of nitric oxide, leading to increased cGMP levels in the corpus cavernosum, resulting in smooth muscle relaxation and inflow of blood to the penis.

Side Effects
Contraindications
- Concomitant use with nitrates (e.g., nitroglycerin) in any form (oral, sublingual, transdermal, injectable) due to potentiation of hypotensive effects.
- Hypersensitivity to Sildenafil or any component of the tablet.
- Concomitant use with guanylate cyclase stimulators like Riociguat.
- Patients with a history of non-arteritic anterior ischemic optic neuropathy (NAION) regardless of previous PDE5 inhibitor exposure.
- Severe cardiovascular disorders (e.g., unstable angina, severe heart failure), recent stroke or myocardial infarction.
Drug Interactions

Excessive alcohol consumption may increase the risk of side effects like dizziness and headache.
Nitrates
Potentiation of hypotensive effects, leading to severe and potentially fatal hypotension. Contraindicated.
Riociguat

In studies with healthy volunteers, single doses up to 800 mg resulted in similar adverse events as seen at lower doses, but with increased incidence and severity. In case of overdose, standard supportive measures should be adopted as required. Renal dialysis is not expected to accelerate clearance as Sildenafil is highly protein bound and not significantly excreted in urine.
