Description
Overview of the medicine
Amoxon 125 mg Suspension is a broad-spectrum penicillin antibiotic used to treat various bacterial infections in children.
Uses & Indications
Dosage
Adults
Not typically used for adults as 125mg/5ml suspension, but adult dosages are usually 250-500 mg every 8 hours or 500-875 mg every 12 hours (using other forms).
Elderly
Dosage similar to adults, but renal function should be considered and adjusted if impaired.
Renal_impairment
Dose reduction or increased dosing interval is required based on creatinine clearance (e.g., CrCl < 30 mL/min, maximum 500 mg every 12 hours).
How to Take
Administer orally. Can be taken with or without food. Shake the bottle well before each use. Use the provided measuring spoon/cup for accurate dosing.
Mechanism of Action
Amoxicillin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Approximately 30 minutes to 1 hour.
Excretion
Primarily excreted unchanged in the urine (50-70%) within 6 hours via renal tubular secretion and glomerular filtration.
Half life
Approximately 1-1.5 hours in adults with normal renal function; longer in neonates and patients with renal impairment.
Absorption
Rapidly and well absorbed from the gastrointestinal tract (75-90%). Food does not significantly affect absorption.
Metabolism
Minimally metabolized in the liver (10-25%).
Side Effects
Contraindications
- Known hypersensitivity to penicillin or any component of the formulation.
- History of severe allergic reaction to other beta-lactam antibiotics (e.g., cephalosporins, carbapenems).
Drug Interactions
Probenecid
Decreases renal tubular secretion of amoxicillin, resulting in increased and prolonged blood levels of amoxicillin.
Allopurinol
Increased incidence of skin rash when co-administered.
Methotrexate
May decrease renal clearance of methotrexate, leading to increased toxicity.
Oral contraceptives
May reduce the efficacy of oral contraceptives in rare cases.
Oral anticoagulants (e.g., Warfarin)
May prolong prothrombin time (INR); monitor closely.
Storage
Store powder below 30°C, protected from light and moisture. Keep out of reach of children. Reconstituted suspension should be stored in a refrigerator (2-8°C) and discarded after the recommended period.
Overdose
Symptoms of overdose include nausea, vomiting, diarrhea, and abdominal pain. In severe cases, crystalluria leading to renal failure has been reported. Management is symptomatic and supportive; hemodialysis may be considered in severe cases.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown harm to the fetus. Amoxicillin is excreted in breast milk in small quantities; generally considered safe during lactation, but monitor infants for potential diarrhea or candidiasis.

Lab Monitoring
- Renal function tests (for prolonged therapy or in patients with pre-existing renal impairment)
- Complete Blood Count (CBC) (for prolonged therapy)
- Liver function tests (if signs of hepatic dysfunction appear)
Doctor Notes
- Educate parents on the importance of completing the full course of antibiotics to prevent resistance.
- Advise on proper reconstitution, storage, and accurate measurement of doses for children.
- Be alert for signs of allergic reactions, especially in patients with a history of penicillin allergy, and advise parents on when to seek immediate medical attention.
- Consider alternative antibiotics if a patient experiences persistent diarrhea, which could indicate C. difficile infection.
Patient Guidelines
- Complete the entire prescribed course of treatment, even if symptoms improve, to prevent resistance and recurrence.
- Shake the suspension bottle well before each dose.
- Store the reconstituted suspension in the refrigerator and discard any unused portion after the recommended period (usually 7 or 14 days).
- Report any signs of allergic reaction (e.g., rash, difficulty breathing) immediately to your doctor.
Missed Dose Advice
If a dose is missed, take it as soon as you remember, unless it is almost time for the next dose. In that case, skip the missed dose and continue with the regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Amoxicillin is generally not expected to affect the ability to drive or operate machinery. However, dizziness (an uncommon side effect) may occur, so caution is advised if affected.
Lifestyle Advice
- Maintain good hydration by drinking plenty of fluids.
- Ensure proper hygiene to prevent the spread of infection.
