Description
Overview of the medicine
Amoxon 250 mg Capsule contains Amoxicillin, a broad-spectrum penicillin antibiotic used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
Typical adult dose: 250 mg to 500 mg every 8 hours, or 500 mg to 875 mg every 12 hours, depending on the infection severity and type.
Elderly
Dosage adjustments may be necessary based on renal function.
Renal_impairment
Dose reduction or increased dosing interval may be required; e.g., for CrCl < 30 mL/min, dosage adjustment is necessary.
How to Take
Oral administration. Can be taken with or without food. For best absorption, it is often recommended to take it at the start of a meal.
Mechanism of Action
Amoxicillin acts by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins (PBPs) located in the bacterial cell wall, which prevents the cross-linking of peptidoglycan polymers, leading to cell lysis and bacterial death.
Pharmacokinetics
Onset
30-60 minutes
Excretion
Primarily excreted unchanged by the kidneys via tubular secretion and glomerular filtration. Approximately 60-80% of a dose is excreted in urine within 6 hours.
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, especially when taken with or without food. Bioavailability is approximately 70-90%.
Metabolism
Limited metabolism, primarily to inactive penicilloic acid.
Side Effects
Contraindications
- Hypersensitivity to amoxicillin or any other penicillin antibiotic
- History of severe allergic reaction to beta-lactam antibiotics (e.g., cephalosporins, carbapenems)
Drug Interactions
Probenecid
Increases and prolongs amoxicillin blood levels by reducing renal tubular secretion.
Methotrexate
May increase methotrexate toxicity due to reduced renal clearance.
Oral contraceptives
May reduce the efficacy of oral contraceptives (rarely significant, but caution advised).
Warfarin and other oral anticoagulants
May increase the anticoagulant effect, requiring monitoring of INR.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include gastrointestinal symptoms (nausea, vomiting, diarrhea) and disturbances of fluid and electrolyte balance. In rare cases, crystalluria or renal impairment may occur. Treatment is generally supportive and symptomatic. Hemodialysis may be used to remove amoxicillin from circulation.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe during pregnancy, but use only if clearly needed. Excreted in breast milk in small amounts; generally considered safe for breastfeeding, but monitor infant for potential adverse effects (e.g., diarrhea, candidiasis).
