Antifungalin is a broad-spectrum antifungal agent used to treat various fungal infections caused by susceptible organisms.
Uses & Indications
Dosage
Adults
The dosage varies based on the infection. For vaginal candidiasis: 150 mg single oral dose. For systemic infections: 200-400 mg once daily. For onychomycosis: 150 mg once weekly.
Elderly
No dose adjustment is typically needed unless there is significant renal impairment. Monitor renal function.
Renal_impairment
Dose adjustment required. For creatinine clearance <50 mL/min, reduce dose by 50% after the initial loading dose.
How to Take
Take Antifungalin capsule orally with or without food. Swallow the capsule whole with a glass of water. Do not chew or crush.
Mechanism of Action
Antifungalin inhibits the synthesis of ergosterol, a vital component of fungal cell membranes, leading to increased membrane permeability and subsequent cell death.

Within 24-48 hours for symptomatic relief, full effect may take several days.

Mainly excreted renally as unchanged drug and metabolites.

Approximately 15-30 hours, allowing for once-daily dosing.

Side Effects
Contraindications
- •Hypersensitivity to Antifungalin or other azole antifungals.
- •Concomitant use with drugs known to prolong QTc interval and metabolized by CYP3A4 (e.g., terfenadine, astemizole, cisapride, quinidine).
Drug Interactions
Warfarin
Increased prothrombin time; monitor INR closely.
Phenytoin
Increased phenytoin levels; monitor levels.
Rifampicin
Decreased Antifungalin levels; consider higher Antifungalin dose.
Cyclosporine
Increased cyclosporine levels; monitor levels.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptoms may include hallucinations, paranoia, and hepatic dysfunction. Management is symptomatic and supportive, possibly involving gastric lavage and forced diuresis. Hemodialysis significantly reduces plasma levels.
Pregnancy & Lactation
Pregnancy Category D (for long-term high-dose use); use only if potential benefit justifies potential risk to the fetus. Small amounts are excreted in breast milk; caution advised during lactation.
